Planting mulberry and raising Silkworm is one of the traditional industries of Chinese agriculture,and different parts of mulberry resources are traditional Chinese herbs.The dried leaves of Morus alba L.are medicated as Mori Folium,which has the effect of evacuating wind heat,clearing lungs and clearing liver.The dried branches of Morus alba L.are medicated as Mori Ramulus,which has the effect of rheumatism and joints.The dried root bark of Morus alba L.are medicated as Mori Cortex,which has the effect of relieving lung and relieving asthma,and diminishing water and swelling.The dried fruit of Morus alba L.are medicated as Mori Fructus,which has the effect of nourishing yin and blood,and moistening and moistening.Does the mulberry source of different parts of mulberry origin have different effects?What is the basis of the pharmacodynamics related to the traditional efficacy of mulberry herbs?What are the similarities and differences in the chemical composition of mulberry medicinal materials,and how does it relate to the efficacy?At present,there have been reported on the chemical composition analysis and pharmacodynamics of some parts of mulberry herbs,but the material basis of the homologous effect of mulberry herbs have not been systematically studied.Therefore,this thesis used the high performance liquid chromatography-mass spectrometry?HPLC-MS?method to systematically study the chemical constituents of mulberry herbs?Mori Folium,Mori Ramulus,Mori Cortex,Mori Fructus?for the first time,and analyzed the chemical composition and similarities between mulberry herbs.The molecular docking technology was used to virtually screen the anti-inflammatory and anti-cough material basis of mulberry herbs,which provided a reference for the quality control of mulberry medicinal materials and the basis for the clinical application of mulberry medicinal materials.The main research contents and results are as follows:1.The analysis of methanol extract of mulberry herbs by HPLC-MS An HPLC-MS technique was used to qualitatively analyze the chemical constituents of the methanol extract of mulberry herbs.A total of 46 compounds were identified,including 5 phenolic acids and 30 flavonoids.4 kinds of Diels-Alder type adducts,2kinds of stilbenes,4 kinds of 2-arylbenzofurans,1 kind of terpenoids.Sixteen compounds were identified in Mori Folium,14 compounds were identified in Mori Ramulus,30 compounds were identified in Mori Cortex and 9 compounds were identified in Mori Fructus.There are 10 kinds of compounds in Mori Cortex and Mori Ramulus,which are Chlorogenic acid,Mulberroside A,Dihydromorin,Oxyresveratrol,Kuwanon G,Sangenone G,Kuwanon K,Cudraflavone C,Kuwanon H and Sanggenol A.There are 9 compounds in Mori Folium and Mori Fructus,which are1-caffeoylquinic acid,Cryptochlorogenic acid,Chlorogenic acid,Neochlorogenic acid,Rutin,Isoquercitrin,Kaempferol-3-O-rutinoside,Astragalin and Quercetin.The common constituent of them is Chlorogenic acid.The exclusive compounds in Mori Folium are Quercetin-3,7-di-O-?-D-glucopyranose,Kaempferol-3,7-di-O-?-D-glucop yranose,Quercetin-3-O-?-D-pyranosyl-?1,6?-?-D-glucopyranoside,6??-Acetylisoquerce tin,6''-Acetyl astragaline,and 6??-O-crotonyl astragaline.The exclusive compounds in Mori Ramulus are Quinic acid and Kuwanon C.The exclusive compounds in Mori Cortex are Morin,Moracin O,Mulberroside C,Kuwanol A,Sangenone T,Kuwanon Y,Mulberrofuran G,Mulberrofuran C,Albanin A,Mulberrofuran J,Limonin,Sangenone C,Kuwanon W,Kuwanon O,Sangenone D,Cyclomulberrin,Moracin,Sanggenol M,Moracin dimer and Kuwanon U.2.The analysis of different polar fractions in water extracts of mulberry herbs by HPLC-MS An HPLC-MS technique was used to qualitatively analyze the chemical constituents of different polar fractions in water extracts of mulberry herbs.A total of 50 compounds were identified,and 7 compounds were identified from petroleum ether.6 compounds were identified from the chloroform site,and 28compounds were identified from the ethyl acetate site.18 compounds were identified from the n-butanol site.There were 4 kinds of compounds in common,which were9,10,11-trihydroxy-12-octadecenoic acid,Arachidic acid,Isoquercitrin and Morin-3-O-?-D-glucopyranoside.The exclusive compounds in Mori Folium are Naringin,Syringin,5-hydroxycoumarin-7-O-?-D-furan glucoside,Moracin M,Kaempferol-7-O-glucoside,Moracin D,Moracin G,Moracin C,Quercetin-3,7-di-O-?-D-glucopyranose,kaempferol-3,7-di-O-?-D-pyridin Glucosamine,Guangsangon L,3'-methoxyquercetin.The exclusive compounds in Mori Ramulus are Cyclomorusin,Cudraxanthone S and Liriodendrin-A.The exclusive compounds in Mori Cortex are Ethyl palmitate,N-heneicosanoic acid,Taxifolin,Morin,Mulberroside C,Moracin O,Scopoletin,Cathafuran B,Kaempferol and Quinic acid.The exclusive compounds of Mori Fructus are 9,12,13-trihydroxy-10-octadecenoic acid,Neochlorogenic acid,Cryptochlorogenic acid,Rutin,Kaempferol-3-O-rutinoside,6''-acetyliso quercetin,Kaempferol-3-O-?-D-pyranosyl-?1,6?-?-D-glucopyranosideandBenzoicacid4-O-?-D-glucopyranoside.Comprehensive analysis of the above HPLC-MS results from mulberry herbs.A total of 78 compounds were identified from them.They are 7 fatty acids,42flavonoids,5 coumarins,9 2-arylbenzofurans,5 Diels-Alder adducts,5 phenolic acids,2 stilbene Class,1 triterpenoid and 2 other compounds.Among them,31 compounds were identified in Mori Folium,27 compounds were identified in Mori Ramulus,47compounds were identified in Mori Cortex,and 22 compounds were identified in Mori Fructus.Among them,Chlorogenic acid,Isoquercitrin,Quercetin,9,10,11-trihydroxy-12-octadecenoic acid,Arachic acid and Morin-3-O-?-D-glucopyranoside are all existed in four mulberry herbs.There are 11 kinds of exclusive compounds in Mori Folium,3 kinds of exclusive compounds in Mori Ramulus,20 kinds of exclusive compounds in Mori Cortex,and 3 kinds of exclusive compounds in Mori Fructus.It can be seen that there are not only common compounds but also specific components among the mulberry herbs.3.Virtual screening of the anti-inflammatory and anti-cough material basis of mulberry herbs based on molecular docking technology Nitric oxide synthase,Tumor necrosis factor-?,Interleukin-6 and Interleukin-1?were used as anti-inflammatory research targets of mulberry herbs,and the anti-inflammatory substance basis of mulberry herbs was screened based on molecular docking.Docking with nitric oxide synthase showed that Cathafuran B,6??-Acetyl astragaline,Quercetin,Astragaline,Benzoic acid 4-O-?-D-glucopyranoside,6??-O-crotonyl astragaline,Cudraflavone C and Sangenone D have potential Nitric oxide synthase inhibition.Among them,4 compounds are in Mori Folium,including6??-Acetyl astragaline,Quercetin,Astragaline and 6??-O-crotonyl astragaline.2compounds are in Mori Ramulus,including Quercetin and Cudraflavone C.4compounds are in Mori Cortex,including Cathafuran B,Quercetin,Cudraflavone C and Sangenone D.2 compounds are in Mori Fructus,including Quercetin and Benzoic acid 4-O-?-D-glucopyranoside.Docking with Tumor necrosis factor-?showed that Cathafuran B,Kuwanon K,Kuwanon Y,Kuwanon O,Mulberrofuran G,Cyclomorusin and Morusin dimer have potential Tumor necrosis factor-?inhibitory activity.Among them,6 compounds are in Mori Cortex,including Cathafuran B,Kuwanon K,Kuwanon Y,Kuwanon O,Mulberrofuran G and Morusin dimer.Cyclomorusin is the exclusive compounds in Mori Ramulus.Docking with Interleukin-6 showed that Cathafuran B,Morusin dimer,Isoquercitrin,and Kuwanon U have potential Interleukin-6 inhibition.Among them,6compounds are in Mori Cortex,including 4 compounds are in Mori Cortex,including Cathafuran B,Morusin dimer,Isoquercitrin,and Kuwanon U.And Isoquercitrin is all in them.Docking with Interleukin-1?showed that Rutin,Quercetin-3-O-?-D-pyranosyl-?1-6?-?-D-glucopyranoside,6??-O-crotonyl astragaline,Cyclomorusin and Kuwanon Y have potential Interleukin-1?inhibitory activity.Among them,3 compounds are in Mori Folium,including Rutin,Quercetin-3-O-?-D-pyranosyl-?1-6?-?-D-glucopyrano-Side and 6??-O-crotonyl astragaline.2 compounds are in Mori Ramulus,including Quercetin-3-O-?-D-pyranosyl-?1-6?-?-D-glucopyranoside and 6??-O-crotonylastrag aline.Kuwanon Y is in Mori Cortex.Rutin is in Mori Fructus.The above results indicate that among the compounds with good anti-inflammatory target molecules in mulberry herbs,there are 6 compounds in Mori Folium,5 compounds in Mori Ramulus,10 compounds in Mori Cortex,and 5compounds in Mori Fructus.Phosphodiesterase 4B and?2 adrenergic receptors were used as cough reference genes for mulberry herbs,and the basis of coughing substances of mulberry herbs was screened based on molecular docking.Docking with phosphodiesterase 4B showed that Quinic acid,Cathafuran B,Kuwanon U,Isoquercitrin,Ethyl palmitate,Sangenone G,Cryptochlorogenic acid and Morusin dimer have potential phosphodiesterase 4B inhibitory activity.Among them,3 compounds are in Mori Folium,including Quinic acid,Isoquercitrin and Cryptochlorogenic acid.3 compounds are in Mori Ramulus,including Quinic acid and Isoquercitrin.7 compounds are in Mori Cortex,including Quinic acid,Cathafuran B,Kuwanon U,Isoquercitrin,Ethyl palmitate,Sangenone G and Morusin dimer.2compounds are in Mori Fructus,including Isoquercitrin and Cryptochlorogenic acid.Docking with?2 adrenergic receptors showed that Kuwanon U,Cathafuran B,Cudraflavone C,Sangenone D,Morusin dimer,Kuwanon W,Limonin and Sanggenol A have potential?2 adrenergic receptor inhibitory effects.Among them,8 compounds are in Mori Cortex,including Kuwanon U,Cathafuran B,Cudraflavone C,Sangenone D,Morusin dimer,Kuwanon W,Limonin and Sanggenol A.Cudraflavone C is in Mori Ramulus.The above results indicate that there are three compounds in the Mori Folium,two compounds in the Mori Ramulus,14 compounds in the Mori Cortex,and two compounds in the Mori Fructus.
