Dendrobium huoshanense is an herbaceous plant of the genus Dendrobium,and it is also a rare species in traditional Chinese herbal medicines of Dendrobium.It has anti-oxidation,anti-fatigue,anti-tumor and pharmacological activities for treating cataract.Malignant tumors are health problems that threaten human life In modern times.Chemical drug therapy is one of the important means to treat tumor diseases and prevent the deterioration of tumor diseases.However,the selectivity of chemotherapy drugs is poor,and they are also seriously damage normal cells that are active in growth while killing tumor cells,and directly injury heart,liver,kidney and nerve and immune system functions.At present,it has been reported that Dendrobium huoshanense has pharmacological effects of liver protection and immunity regulation,but there is no literature report on the attenuation effect of Dendrobium huoshanense on liver damage and immunosuppression caused by chemotherapy drugs.Therefore,this study used Dendrobium huoshanense as the research object,and used the chemotherapy representative drug cyclophosphamide to establish liver injury and immunosuppression models to investigate the hepatoprotective activity and immune regulation of Dendrobium huoshanense,respectively,then to evaluate the attenuation effect of Dendrobium huoshanense on cyclophosphamide toxicity,and provide experimental basis for the rational application of Dendrobium huoshanense.In addition,the subject also studied the effects of Dendrobium huoshanense on the activities of cytochrome P450 isoforms CYP1A1,CYP1A2 and CYP2 B,and provided reference for clinical safe drug use of Dendrobium huoshanense.The main results are as follows: 1.Protective effect of Dendrobium huoshanense on liver injury induced by cyclophosphamide The liver injury model of C57BL/6 mice was established by using the cyclophosphamide,which was commonly used chemotherapy drug,to evaluate the hepatoprotective effect of Dendrobium huoshanense.The results showed that Dendrobium huoshanense could reduce the increased levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and malondialdehyde(MDA)in serum caused by cyclophosphamide,and increase the activity of superoxide dismutase(SOD)and alleviate the pathological damage of liver tissue.The results indicated that Dendrobium huoshanense can alleviate liver damage caused by CPA and play a certain role in liver protection.2.Effect of Dendrobium huoshanense on immunosuppression induced by cyclophosphamide The immune injury model was established by intraperitoneal injection of cyclophosphamide to evaluate the immunomodulatory effects of Dendrobium huoshanense.The results showed that Dendrobium huoshanense could improve the reduce of the serum hemolysin level,carbon particle clearance index,DTH ear swelling and cytokine IL-2,IL-6 and TNF-? mRNA expression induced by cyclophosphamide injection.In addition,after oral administration of Dendrobium huoshanense,the body weight of the mice and the percentage of peripheral blood leukocytes,red blood cells,hemoglobin and lymphocytes increased to some extent.The results indicated that Dendrobium huoshanense has a good regulatory effect on the immune function,and it has a protective effect on immune function damage caused by cyclophosphamide,which may be related to the cytokines involved in the regulation of the pathway involved in immune regulation.3.Effect of Dendrobium huoshanense on CYP450 Enzyme Activity in Mice Liver The specific probe substrate method was used to evaluate the effects of Dendrobium huoshanense on the activities of CYP1A2,CYP1A1 and CYP2 B.By comparing in vitro metabolism of the probe substrates phenacetin,bupropion and ethoxylated oxazolone,respectively with the liver microsomes from Dendrobium huoshanense group mice and the blank control group mice,it was found that Dendrobium huoshanense could significantly induce the activity of CYP1A2 enzyme,and no significant effect was found on the activity of CYP2 B and CYP1A1 enzymes.Therefore,it is necessary to pay attention to the effect of Dendrobium huoshanense on CYP1A2 enzyme,and to avoid the rational drug effect of the clinical drugs,which could metabolize by CYP1A2,during combing with Dendrobium huoshanense,The long-term use of it as a health care drug has no adverse effects on the human body.In summary,Dendrobium huoshanense can be used as a long-term health care medicine.Dendrobium huoshanense,while combining with cyclophosphamidem,can protect liver damage and immune function damage caused by cyclophosphamide.
