Purpose:To study the effect of Pidan Ning on apoptosis of islet β cells and expression of protein Fas in islet β cellsfrom rats with impaired glucose regulation.Materials and methods:20rats were randomly selected from 100 rats as blank group,and the remaining 80 rats were fed with high calorie diet.After 12 weeks,all rats were weighed and the oral glucose tolerance test was conducted.The body weight,fasting blood glucose and oral glucose tolerance test 2 hours plasma glucose of rats were recorded.Then the rats with impaired glucose regulation were checked and randomly divided into model group,Pidan Ning group and Metformin group.6 rats in each group were randomly selected to observe the levels of fasting insulin,insulin levels after first phase and second phaseby high glucose clamp technique.The Pidan Ning group and Metformin group were given corresponding drug for 4 weeks.During the same period,the blank group and model group were given distilled water.After the medicine intervention period,6 rats which randomly selected from blank group,and the remaining rats from model group,Pidan Ning group and Metformin group,were measured the body weight,fasting blood glucose and oral glucose tolerance test 2hours plasma glucose.And the levels of fasting insulin,insulin levels after first phase and second phase of rats were observed by high glucose clamp technique.Pancreas of rats were taken after high glucose clamp technique,the number of apoptotic islet βcells and the expression of Fas protein in islet β cells were observed by TdT-mediated UTP nick-end labeling,Immunohistochemical method.Results:1.Weight: After medicine intervention,the body weight of rats in Pidan Ning group and Metformin group was remarkably lower than the body weight of rats in model group(P < 0.01).2.Fasting blood glucose: After medicine intervention,fasting blood glucose of rats in Pidan Ning group was remarkably lower than fasting blood glucose of rats in model group(P < 0.05),while fasting blood glucose of rats in Metformin group was moresignificant(P < 0.01).The oral glucose tolerance test2 hours plasma glucose of rats in Pidan Ning group and Metformin group was remarkably lower than oral glucose tolerance test2 hours plasma glucose of rats in model group(P < 0.01).3.Islet β cell secretion function: After medicine intervention,fasting insulin of rats in Pidan Ning group and Metformin group was remarkably lower than fasting insulin of rats in model group(P < 0.01),and the levels of insulin after first phase and second phase of rats in Pidan Ning group were remarkably higher than levels of insulin after first phase and second phase of rats in model group(P < 0.05),while levels of insulin after first phase and second phase of rats in Metformin group was more significant(P< 0.01).4.Apoptosis of islet β cells: After medicine intervention,the number of apoptotic islet β cells of rats in Pidan Ning group and Metformin group was remarkably lower than the number of apoptotic islet β cells of rats in model group(P < 0.01).5.The expression of protein Fas in islet β cells: After medicine intervention,the optical density mean of islet β cells of rats in Pidan Ning group and Metformin group was remarkably lower than optical density mean of islet β cells of rats in model group(P < 0.01).The optical density mean of islet β cells of rats in Pidan Ning group was higher than optical density mean of islet β cells of rats in blank group,but not significant(P > 0.05).Conclusion:1.Pidan Ning can remarkably reduce the body weight of rats with impaired glucose regulationmodel.2.Pidan Ning can effectively reduce fasting blood glucose and oral glucose tolerance test 2 hours plasma glucose in rats withimpaired glucose regulation.3.Pidan Ning can remarkably improve the function of islet β cells of rats with impaired glucose regulation.4.Pidan Ning can remarkably inhibit the expression of Fas,a kind of protein in islet βcells from rats with impaired glucose regulation,and thus inhibit the apoptosis of islet β cells.
