Purpose:Observation of astragalus membranaceus Huangqi Qianzhong decoction contains medicinal serum combined cisplatin on A549 lung cancer cell proliferation inhibition and TGF-β1,Smad3,Smad7 protein expression changes,discusses the root of remembranous milk vetch chienchung soup combined cisplatin to inhibit cellular proliferation and metastasis of lung cancer related mechanisms,France for ridging raw gold medicine astragalus chienchung soup provide theoretical experiment based on the inhibition of lung cancer cell proliferation.Material and method:The serum containing drugs was prepared by serum pharmacology method,and the rat equivalent dose was calculated by the equivalent dose ratio table of human and animal surface area conversion.Set up the root of remembranous milk vetch chienchung soup of high,medium and low dose groups,respectively,for the adult recommended daily dosage of 2,1,1/2 times,traditional decoction,filtering,the evaporation of water to the crude drugs containing 2.295g/ml,1.148g/ml,0.573g/ml,loaded into sterilization medicine bottles,cooling preserved 4℃.Will only 40 SD rats by randomized controlled method is divided into four groups（male and female half）,respectively,the root of remembranous milk vetch chienchung decoction high dose group,astragalus membranaceus chienchung soup dose group,low dose group and astragalus membranaceus chienchung soup blank serum group,high,medium and low dose of astragalus membranaceus chienchung soup18.36 g/kg,9.18 g/kg,4.59 g/kg and the amount of normal saline lavage,2 times a day,every time 1ml,7d in a row,the last for 1 h abdominal aortic blood,2000 r/min,the centrifugal 5min,take supernatant,56℃water bath 30 min inactivated,0.22μm microporous filtration membrane filtration,-20℃keep aside.Respectively determined by MTT method are used to calculate 10%,5%of high,medium and low dose and high,medium and low dose astragalus chienchung soup medicated serum on human lung adenocarcinoma A549 cell line 24h,48h,72h half inhibitory concentration(IC₅₀),astragalus membranaceus Huangqi Qianzhong decoction soup best drug-containing serum（10%of dose,48h）joint best cisplatin（5μmol/ml,48h）on the strain proliferation inhibition rate;The expression of TGF-β1,Smad3 and Smad7proteins in lung cancer A549 cells was detected by immunohistochemistry.Western Blot analysis of TGF-β1,Smad3 and Smad7 protein expression.Results:1.The IC₅₀0 of A549 cell line treated by serum containing Huangqi Qianzhong decoction was10%medium dose.The inhibition rate of A549 cells in the combined group was higher than that in the traditional Chinese medicine group,the western medicine group and the blank group（P<0.05）.2.Immunohistochemistry showed that,compared with the blank group,TGF-β1 and Smad3protein expression in the TCM group,the western medicine group,Chinese traditional medicine group and the combination group had fewer brown-yellow particles.Compared with western medicine group,TGF-β1 and Smad3 proteins in the combination group had the fewest brown-yellow particles.Compared with the blank group,Smad7 protein expression in TCM group,western medicine group and combination group had more brown-yellow particles.Compared with the western medicine group,the Smad7 protein in the combination group had the most brown-yellow particles.3.Compared with the blank group,TGF-β1 and Smad3 protein expressions were down-regulated in the traditional Chinese medicine group,the western medicine group and the combination group,and the differences were statistically significant（P<0.05）.Compared with western medicine group,TGF-β1 and Smad3 protein expressions were the lowest in the combination group,and the difference was statistically significant（P<0.05）.Compared with the blank group,Smad7 protein expression was up-regulated in the traditional Chinese medicine group,the western medicine group and the combination group,and the difference was statistically significant（P<0.05）.Compared with western medicine group,Smad7protein expression was the highest in the combination group,and the difference was statistically significant（P<0.05）.Conclusion:1.Huangqi Jianzhong decoction combined with cisplatin can inhibit the proliferation of lung cancer A549 cells.2.Huangqi Jianzhong decoction combined with cisplatin can up-regulate the expression of smad7 protein in A549 cells of lung cancer,and down-regulate the expression of TGF-β1 and Smad3 protein.3.These results suggest that Combined group（huangqi jianzhong decoction combined with cisplatin group）can inhibit the proliferation and metastasis of human lung adenocarcinoma A549 cells to some extent,which may be related to the expression of TGF-β1,Smad3 and smad7 proteins in the TGF-β/Smad signaling pathway.
