| EXPRESSION AND CLINICAL SIGNIFICANCE OF GLI-1 AND MMP11 IN HEPATOCELLULAR CARCINOMAObjectivesDiscusses the hepatocellular carcinoma tissues and MMP11 Gli-1 protein expression level,and study them with hepatocellular carcinoma(HCC)the relationship between the clinical classification and pathological factors,for the treatment of hepatocellular arcinoma in clinic to find new breakthrough.MethodsUsing immunohistochemical methods 32 cases of liver cell tissue adjacent to carcinoma tissue and its corresponding 32 cases(from the tumor edge 3 cm or highe-r)and 11 cases of normal liver tissues and MMP11 Gli-1 protein expression level.Results1.The positive expression rate of gli-1 in hepatocellular carcinoma was 71.9%,12.5% in the adjacent tissues,and 9.1% in normal liver tissue,and there was statistically significant difference between groups(P<0.05).Gli-1 expression was significa-ntly lower than that in hepatocellular carcinoma(P<0.05),and there was no signify-cant difference in the expression of gli-1 in adjacent tissues and normal liver(P > 0.05).2.Gli-1 positive expression rate in Ⅰ-Ⅱ period of 40.0% is far less than 86.4% of the Ⅲ-Ⅳ period(P < 0.05),50.0% of the T1 and T2 stage is far less than 85.0% of the T3 and T4 without lymphatic metastasis(P<0.05),46.2% is far less than 89.5% of lymph node metastases(P<0.05),no distant metastasis is far lower than the 45.5% 85.7% of distant metastasis,but its expression and age,sex,whether patients with hepatitis b,whether for liver cirrhosis,preoperative AFP level has nothing to do(P >0.05).3.MMP11 in hepatocellular carcinoma(HCC)positive expression rate of 65.6%,in the tissue adjacent to carcinoma was 18.8%,in the normal liver tissue was 18.2%,thus each expression between the difference was statistically significant(P < 0.05).4.MMP11 protein positive expression rate in Ⅰ-Ⅱ period of 36.4% is far lower than the Ⅲ-Ⅳ period of 81.0%(P<0.05),33.3% of the T1 and T2 stage is far less than 78.3% of the T3 and T4 without lymphatic metastasis(P<0.05),33.3% is far less than 85.0% of lymph node metastases(P<0.05),no distant metastasis-is far lower than the 30.0% 81.8% of distant metastasis,but its expression and age,sex,whether patients with hepatitis b,whether for liver cirrhosis,preoperative AFP level has nothing to do(P>0.05).5.The expression of gli-1 and MMP11 in hepatocellular carcinoma was positiveeely correlated.Conclusion1.Gli-1 is associated with infiltration and metastasis of hepatocellular carcinoma.2.MMP11 is associated with infiltration and metastasis of hepatocellular carcinoma.3.Gli-1 and MMP11 are involved in the occurrence and development of hepatocellular carcinoma,and the higher expression of gli-1 and MMP11 may be one of t-he important factors in promoting infiltration of hepatocellular carcinoma.The joint detection of gli-1 and MMP11 protein expression can provide a theoretical basis for clinical staging and treatment of hepatocellular carcinoma.Section Two GLI-1 AND MMP11 EXPRESSION IN HEPATOCELLULAR CELLObjectivesDetection of human hepatocellular carcinoma cell line SMMC-7221 in the expression of Hedgehog signaling pathways,inhibit the ring dopamine signaling pathwayys after are used to observe Gli-1 MMP11 change,study whether to have a certa-in correlation between the two testing human liver cancer cells and after the interve-ntion ring dopamine Gli-1 MMP11 expression level,explore the Hedgehog and E-MT in correlation of liver cancer cells,and verify the therapeutic effect of ring dopamine.Methods1.Cultured human hepatoma cells SMMC-7221 cells were treated with drug administration.2.MTT assay was used to detect the time-dose-effect curve and medium-effect concentration of cyclopamine on hepatocellular carcinoma cells.3.Western-blot method was used to detect the expression level of Gli-1 MMP11 protein before and after cyclopamine in human hepatocellular carcinoma cell SMMC-7221.Results1.After the treatment of SMMC-7221,the cell proliferation and growth were inhibited.The cells from the full spindle shaped wall grew into a round cake shape,and the inhibition rate increased with the intervention time at a certain concentration.In a considerable range,the drug action time and concentration dependence were in a considerable range.The IC50 after the action of 48 h was 16±0.52μmol/L.2.The Gli-1、MMP11 expression was down-regulated after SMMC-7221.Conclusions1.cyclopamine can inhibit the proliferation of human hepatocellular carcinoma cells SMMC-7221.2.The Hedgehog signaling pathway may be associated with MMP11.3.Cyclopamine may have therapeutic effect on liver cancer. |
