Objective:By observing the effects of the light-colored five kinds of flowers Chinese medicine monomers on the proliferation rate,melanin content and tyrosinase activity of B16F10 melanoma cells cultured in vitro in mice,the mechani sm of flowers Chinese medicine treatment of pigmented diseases was discussed Based on the basic theory,the "Treat Color by Color" and "Treat Skindisease by Flower" methods are initially verified to promote the research and develop ment of traditional Chinese medicines for the treatment of pigmented diseases.Methods:(1)The CCK-8 method was used to detect the inhibition rate of saff lower yellow,quercetin,β-ionone,forsythiaside,and Plumbagin on mouse B16F10 melanoma cells.(2)The NAOH lysis method was used to determine the changes of the melanin content in the cells of the five kinds of Chinese herbal me dicines after low,medium and high concentrations of 48 hours on B16F10 cell s.(3)Advanced Enzyme method was used to detect the effect of 5 kinds of flowers Chinese medicine monomers on tyrosinase activity of B16F10 cells.Results: 1.Safflower yellow,quercetin,β-ionone,forsythiaside,and Plumbagin have different degrees of inhibition on mouse B16F10 melanoma cells(p<0.05),And in a certain range of concentration has a concentration dependence.Compared with the negative control group(p>0.05),the inhibitory effect of low concentration of safflower yellow pigment and white flower saliva on the cells was low,and the cytotoxicity was low.Compared with the positive control g roup,the medium and high concentrations of β-ionone,forsythiaside,and quercetin showed a statistically significant difference(p<0.05).Quercetin,β-ionone,and forsythiaside solutions at low,medium,and high concentrations had a statistically significant difference in cell inhibition rate compared with the negative control group(p<0.05).3.The monomer solution of five kinds of flowers of traditional Chinese medicine can inhibit the activity of tyrosinase in cells,and the inhibition rate also increases with the increase of concentration.The safflower yellow solution(50 ug/ml ~ 250 ug/ml)had statistically significant differe nce in the inhibition rate of cell tyrosinase activity compared with the negative control group(p<0.05).Compared with the positive control group,the high concentration of safflower yellow was statistically significant(p<0.05).Quercetin(2 ug/ml~50 ug/ml),β-ionone,forsythiaside,and Plumbagin(10 ug/ml~50 ug/ml)on B16F10 cell tyrosinase activity The difference between the inhibitory effect and the negative control group was statistically significant(p<0.05).The high concentration of safflower yellow,β-ionone,forsythiaside and Plumbagin solution on B16F10 cell tyrosinase activity was significantly different from the positive control group(p<0.05).Conclusion:1.The light-colored floral traditional Chinese medicine ingredients,mainly composed of safflower yellow,quercetin,β-ionone,forsythiaside,and Plumbagin,have effects on melanoma cell proliferation,melanin content and tyrosine Acidase activity has different degrees of inhibition.2.The theory of “treating color with color” has a certain scientific basis: the light-colored flowers of traditional Chinese medicine have an inhibitory effect on melanin metabolism.3.The safflower yellow and Plumbagin have low toxicity to B16F10 melan oma cells of mice,and have a certain inhibitory effect on melanin metabolism and tyrosinase activity.In the future,they can be used as one of the traditional Chinese medicine monomers with whitening effect.
