Objective:Establish high loads mycoplasma pneumoniae(MP)infection of BALB/c mice severe mycoplasma pneumonia(SMPP)model,the application of nourishing Yin clearing lung soup intervention in severe mycoplasma pneumonia in mice and explore Yangyin Qingfei Tang of high loads induced by MP SMPP BALB/c mice model of NLRP3 inflammatory corpuscle and related factor expression and the effect of further clarify Yangyin Qingfei Tang intervention of SMPP targets.Material and method:96 BALB/c mice were randomly divided into normal group,model group,azithromycin group,Yangyin qingfei soup flavor group,and were named group A,group B,group c,and group D,respectively.With the exception of group A,the SMPP model of mycoplasma pneumoniae(MP)infection was established by continuous drip of MP bacterial solution with A high load into the lower respiratory tract,followed by drug intervention.3 days,7 days after infection,10 days,14 days,make,each random death 6 mice,using HE staining to observe the lung pathology in mice,mice lung tissue ultrastructure with transmission electron microscopy(sem)observation,using Western blotting method to detect lung tissue NLRP3,mottled of apoptosis related proteins(ASC),aspartic acid proteolytic enzymes containing cysteine-1(caspase 1)protein expression,The levels of interleukin-1serum(il-1)and reactive oxygen species(ROS)in serum and alveolar lavage solution were detected by ELISA and the general status of mice was observed.Results:Results:1 After modeling,mice in the high-load group showed reduced activity,reduced diet,weight loss,and increased secretions.After dissection,pathological changes such as hyperemia,consolidation,and suppuration were found in the lung tissue of the mouse.Partially destroyed,there were a lot of inflammatory cell infiltration,and even the alveoli were completelyconsolidated.Compared with the normal group,the two model groups had higher lung index and pathological score at each time point than the normal group(P <0.05).On days 7,7,and10,the lung index and pathological score of the high-load group were higher than those of the normal-load nasal drip group(P <0.05),indicating that the model was successfully established,and it was found that the mice of the two model groups tended to be stable after 14 days.2 After transmission electron microscopy scanning,it was found that the lung tissues of MP-infected mice showed obvious destruction of alveolar epithelial cells and cell proliferation,and the microvilli of the free surface of type Ⅱ alveolar cells thickened and fell off,uneven distribution of chromatin and small plates The body is emptied and vacuolated.In the azithromycin group,the microvilli were slightly thickened,a small amount fell off,and the arrangement was uneven,and some mitochondria were swollen.In the Yangyinqingfeitang Jiawei group,the microvilli fell off a little,and the arrangement was still neat.3 After HE staining,it was found that the bronchiole wall of the model group was thickened,the alveolar structure of the mouse was seriously damaged,the alveolar interval was significantly widened,the tracheal epithelium was shed,a large number of inflammatory cells oozed out,and the capillaries of the lung tissue were congested;the azithromycin group was small Compared with the model group,the number of inflammatory cells was significantly improved,and only a small amount of inflammatory cell infiltration was observed;the mice in the Yangyin Qingfei Tang Jiawei group had only a small amount of inflammatory cells in the alveoli and moderate congestion of lung tissue.After infection,the lung index and pathological score were higher than those in the normal group(P <0.05),and tended to peak on the 7th day.4 Western blot method to detect NLRP3,ASC,caspase-1,ELISA method to detect ROS,IL-1β results showed that compared with the normal group,NLRP3,ASC,caspase-1,3,7,10,14 days after MP infection,ROS and IL-1β expressions increased(P <0.05)and reached a peak on the 7th day.After drug intervention,compared with the normal group,the expression of NLRP3,ASC,caspase-1,IL-1β in the two-dose group was higher than that in the normalgroup;compared to the model group,NLRP3,ASC,caspase-1.The expression of IL-1βwas all down-regulated(P <0.05).Among them,the expression of NLRP3 inflammatory bodies was lower than that of the azithromycin group after the drug intervention in the Yangyinqingfeitang decoction group(P <0.05).Conclusion:1.A mouse model of severe mycoplasma pneumoniae pneumonia can be established by nasal drops with a high load of MP bacteria2.Yangyin qingfei soup flavor can effectively reduce lung tissue inflammation in SMPP mice.3.Yangyin qingfei soup flavor can effectively down-regulate the expression levels of ROS,NLRP3,ASC,caspase-1 and serum il-1 in the lung tissues of mp-infected mice,which may be a target for the treatment of SMPP.
