| Purpose:In this study,Liao Sakamoto was used to intervene as rabbits in animal experiments.By detecting the levels of cytokines TNF-α,IL-6,IL-8 and IL-10 in the serum of atherosclerotic rabbits,we studied whether Liao Sakamoto affected the immune balance of Th1 / Th2 by regulating the level of cytokines to explore its mechanism in the treatment of atherosclerosis.Material and method:50 male Japanese white rabbits were randomly divided into 2groups: 10 in the blank group and 40 in the model group.After 7 days of adaptive feeding,the blank group continued to receive normal feed feeding,and no intervention was performed.The model was made for high-fat feeding,and the carotid balloon injury was performed 4weeks after the module was formed.After the model was built,the blank group and the model were randomly selected to determine whether the model was successful.After the model was successfully built,Eight rats were randomly selected from the model group four weeks after operation to determine whether the model was successful.After the model was successful,the model group was randomly divided into two subgroups: model group and Liao Sakamoto group..The sputum group was intragastrically administered once a day,and the dose was0.38 g/kg;the model group was given daily distilled water.After 4 weeks of treatment,blood was taken from the ear vein,and carotid tissue was taken after anesthesia.HE staining was used to detect the histological changes of carotid atherosclerotic plaque in rabbits.The serum was determined by enzyme-linked immunosorbent assay(ELISA).TNF-α,IL-6,IL-8,IL-10content;detection of serum triglyceride(TG),total cholesterol(TC),high density lipoprotein(HDL),low density lipoprotein(LDL)content.Results:1.Compared with the blank group,the TG,TC,HDL,LDL of the model group was significantly higher than that of the blank group(P < 0.05).After the intervention of Liao Sakamoto,the TG,TC,HDL,LDL of Liao Sakamoto group was lower than that of the model group,and the results were statistically significant(P < 0.05).2.The effect of sputum on themorphology of AS rabbits2.The effect of Liao Sakamoto on the histomorphology of AS rabbits in the blank group,the carotid artery thickness was uniform,the intima was smooth,the structure was intact,and the endothelial cells were not damaged.The thickness of arterial wall in the model group was different and uneven.Obvious atherosclerotic plaques were formed.Endothelial cells were damaged,exfoliated,intimal hyperplasia,and a large number of foam cells and a small number of cell fragments could be seen under the intima.A small number of fibrous plaques were observed and the accumulation of foam cells under the membrane was significantly decreased in Liao Gao group.3.The effect of Liao Sakamoto on serum TNF-α,IL-6,IL-8,IL-10 in as rabbits.The results of serum ELISA showed that TNF-α,IL-6,IL-8 were significantly increased(P <0.05)and IL-10 was significantly decreased(P <0.05)in the model group compared with the blank group.Conclusion:1.Liao Sakamoto.could can effectively improve the histomorphology of carotid plaque in as rabbits.2.Liao Sakamoto.could effectively improve the dyslipidemia in AS rabbits.3.Liao Sakamoto.could regulate the expression of TNF-α and IL-6,IL-8,IL-10 in serum of AS rabbits and interfere with Th1/Th2 equilibrium axis.4.Liao Sakamoto.could have affected atherosclerosis in rabbits by regulating immunity. |
