The incidence of male infertility is increasing year by year.The main reason is oligoasthenospermia.Modern medicine believes that oligoasthenospermia is closely related to environment,endocrine,immunity,infection,varicocele and other factors.Recent studies have found that obesity is also one of the causes of oligoasthenospermia.Obesity belongs to phlegmdampness constitution in TCM constitution classification.Professor Wang Qi put forward that the core pathogenesis of oligoasthenospermia is "kidney deficiency or damp heat and stasis or toxin or insects",while obese people eat more fat,sweet and thick taste,damage gastrointestinal function,gather dampness and generate phlegm,phlegm and dampness stagnation for a long time will become heat,which is consistent with the pathogenesis of "dampness and heat".Therefore,according to the theory of body disease and the thought of preventing disease,this experiment establishes a rat model of obesity oligoasthenospermia,and preventively gives Huatan Qushi Decoction(HTQSD),observes the regulating effect of this recipe on lipid metabolism and sperm parameters of obesity oligoasthenospermia rats induced by high fat,and explores its possible mechanism.Objective: To establish a rat model of obese oligoasthenospermia,and to observe the regulating effect of HTQSD on lipid metabolism and sperm parameters in obese oligoasthenospermia rats induced by high-fat diet,and explore its possible mechanism.METHODS: 8-week-old Wister male rats were randomly divided into blank group,model group,WZYZP group and HTQSD group with 24 rats in each group.The blank group was fed with normal diet,while the other groups were fed with high-fat diet for 12 weeks.The blank group and model group were given the same volume of saline,WZYZP group was given Wuzi Yanzong pill(1.07g/kg),the other groups were given low,medium and high doses of Huatan Qushi Decoction(6.56g/kg,13.13 g/kg,26.25g/kg),the volume of gastric perfusion was 1ml/100 g.In experiment 1,the weight of rats was weighed,and the heart,liver and testis tissues were taken.The sperm of epididymis tail was collected by diffusion method,and the sperm quality was analyzed by blood cell counting plate.In experiment 2,serum lipid metabolism and sex hormone levels were measured by ELISA.In experiment 3,rat testis was stained with HE,and histomorphology was observed by optical microscopy.The pathological changes of spermatogenic cells in testicular seminiferous tubules were observed,and Johnsen score was recorded.In experiment 4,the expression of SOD1 and HO-1 in serum was detected by RTPCR and Western Blot respectively.Results: Compared with the blank group,the body weight,Lees index and liver index of the model group increased significantly at the 8th week(P<0.01,P<0.001),semen concentration and sperm motility rate(P<0.01,P<0.001),serum levels of TG,TC,LDL-C increased significantly(P<0.001),HDL-C decreased significantly(P<0.001),T,E2,LSH decreased significantly(P<0.01,P<0.001),and Johnsen score decreased significantly(P<0.001).The expression of SOD1(superoxide dismutase)and HO-1(heme oxygenase-1)decreased significantly(P<0.001).By the end of the 12 th week,the above indexes were still different,and the testicular index decreased(P<0.05).Compared with the model group,the body weight and Lees index of HTQSD low,medium and high dose groups decreased significantly(P<0.01,P<0.001),the sperm motility rate increased significantly(P<0.01,P<0.001),and the semen concentration of WZYZP and HTQSD medium dose groups increased significantly(P<0.01,P<0.001).In terms of blood lipid,the indexes of TG,TC,HDL-C,LDL-C and T,LSH in HTQSD medium dose group were different at the 4th week.(P<0.01,P<0.001)In WZYZP group,the TG level decreased significantly only at the 8th week(P<0.01),while the TC,HDLC and LDL-C indexes did not change significantly(P>0.05).The T and FSH levels increased at the 4th week and lasted until the 12 th week(P<0.001),while the E2 level increased significantly at the 12 th week(P<0.01).Compared with the model group,the Johnsen scores of WZYZP group and HTQSD medium dose group were different from the 8th week(P<0.01,P<0.001).HTQSD significantly increased SOD1 expression in serum of rats at 12 weeks(P<0.01),but WZYZP had no significant effect.From the 8th week,medium dose HTQSD could increase HO-1 expression level(P<0.01)and lasted for 12 weeks(P<0.001).WZYZP did not show any difference with model group until 12 weeks(P<0.05).Conclusion: HTQSD can prevent the regulation of blood lipids in rats,and it may improve the serum anti-oxidation,reduce the degree of oxidative stress,and improve the semen quality of rats with less prolific sperm.
