Full-field optical coherence tomography(FFOCT)is a novel in-vivo biological tissue optical tomography method with non-invasive and high-resolution features.In this thesis the diagnosis value of en face images obtained by FFOCT and the method of using FFOCT to achieve dynamic contrast imaging.The main contents and results are as follows:By comparing the en face images obtained by FFOCT with the images obtained from histologically stained sections,the main structural features were identified,including renal corpuscle,colonic gland and goblet cells,hepatocytes in the hepatic lobules,central veins,and esophageal epithelial cells.To verify the ability of FFOCT to distinguish between normal and cancerous tissues,en face images of three adenocarcinomas were generated,including colon adenocarcinoma,hepatocellular carcinoma and esophageal tumor.Combined with histopathological slice images,the comparative analysis of the pathological structure and normal structure of these three cancerous tissues was given.Important cellular structure information is often covered by strong scattering structure signals when imaging with FFOCT.To solve this problem,the dynamic contrast imaging method of FFOCT was investigated.The dynamic intensity image of in-vivo tissue was obtained to provide additional contrast for the traditional FFOCT images.The signal model of dynamic FFOCT was derived from the basic single-particle dynamic signal model.It was shown that SVD decomposition can be used to approximately filter out information comes from strongly scattering low activity structures,which enables dynamic contrast imaging with traditional FFOCT systems.In-vivo imaging analysis of human finger skins was performed.The results of this research laid the foundation for the application of FFOCT system to the analysis of en face images of living tissue imaging.
