Astragalus Radix is the dried root of Astragalus membranaceus(Fisch.)Bge.var.mongholicus Bge.Hsiao or Astragalus membranaceus(Fisch.)Bge.[1].It has various functions such as regulating immune function,anti-fatigue and anti-aging,and has been widely used in the clinic.Because of the specific growing environment and cultivation method,Zizhou Astragalus has a gradually increasing influence.Therefore,to prevent the misuse of inferior astragalus and to scientifically and standardizedly match different market demands,how to accurately and identify Zizhou Astragalus,and formulate a reasonable quality grade standard for Zizhou Astragalus is our urgent problem to be solved.The purpose and significance of this project are:(1)Distinguish Zizhou Astragalus from other varieties,find the differentiation characteristics of Zizhou Astragalus,and realize the identification of Zizhou Astragalus.(2)Study about quality differences in different commodity grades based on trait characteristics of Zizhou Astragalus decoction pieces,and determine the rationality of the current commodity-grade standards.(3)Explore quality differences in different commodity grades according to active components of Zizhou Astragalus decoction pieces,and discuss the rationality of the current commodity-grade standards.(4)Draft quality standard for Zizhou Astragalus.The research methods:(1)Experimental methods ① The spectrophotometer,electronic tongue,texture analyzer,electronic balance,tape measure,and vernier caliper were used to measure the color,taste,hardness,mass density,the proportion of xylem and other properties respectively.② The contents of the flavonoids,astragaloside,total flavonoids,total polysaccharide,and other medicinal components were determined using HPLC-PDA,HPLC-ELSD,UV methods.(2)Data processing methods①Determine the multiple trait indexes and effective component indexes of astragalus from different origin places,use Fisher discriminant analysis and stepwise discriminant analysis,and finally achieve the reliable identification of Zizhou astragalus.② Based on the determination of various trait indexes and efficacy component indexes of Zizhou Astragalus decoction pieces,the quality grade division and the screening of grading indexes of Zizhou Astragalus decoction pieces were realized by using principal component analysis,K-means clustering analysis,correlation analysis,and other methods,and the Fisher discriminant analysis,partial least squares regression analysis and other methods are used to construct a quality grade prediction model.Research results:(1)Taking samples with a diameter of 0.5-0.7 cm,determine the L*value and b*value of the powder color of the sample to realize the identification of Zizhou Astragalus.(2)In the seven different commodity grades of Zizhou Astragalus decoction pieces of Gl-G7,the powder color is not entirely positively correlated with the commodity-grade.(3)Among the seven assorted Zizhou Astragalus pieces of G1-G7,only the content of Calycosin-7-glucoside,ononin,and total flavonoids were positively correlated with the commercial-grade,and the other components were negatively correlated or had no clear rules.(4)The four indexes of diameter,b*value,mass density,and calycosin content,are selected as the evaluation indexes of quality grade division.The sample is divided into three grades,and the HQI,constructed quality discrimination index,is used as the index for the prediction of the quality grade.The draft of the quality grade standard for Zizhou Astragalus decoction pieces is finally achieved.Conclusion and significance:(1)Use a spectrophotometer to determine the color of the sample powder to realize the rapid identification of Zizhou Astragalus,which is easy to operate and low in cost.(2)The current commodity-grade standards using the diameter as the main grading-index are unreasonable,and it is necessary to establish new quality grade standards.(3)In this study,the various trait indicators recorded in ancient Chinese herbal medicine classics were fully considered and combined with the pharmacodynamic composition form modern experimental research.Therefore,the samples’ classification and grading are reasonable,and the selected indicators have strong operability.All above indicates that the proposed quality standard for Zizhou Astragalus is more scientific than traditional standards.
