Study On The Spermatogenesis And Molecular Mechanism Of Wuzi Yanzong Pills

In recent years,the incidence of male infertility has been on the rise.Wuzi Yanzong Pills is a classic Chinese medicine treatment of oligozoospermia.It has extensive clinical applications in the treatment of male infertility and is known as the first seed of ancient and modern seeds.Treatment of male infertility and improvement of male sperm quality have become an urgent social problem.However,due to the multi-component and multi-target of traditional Chinese medicine,the mechanism of action of Wuzi Yanzong Pills is not yet clear.Therefore,this study first established a mouse model of Buxuanan caused oligozoospermia,and gave the model animal Wuzi Yanzong Pills through the testis tissue Pathological analysis,sperm quality and fertility evaluation of the spermatogenic effect of Wuzi Yanzong Pills.On this basis,further application of gene chip technology to analyze the effect of Wuzi Yanzong Pills on the gene expression profile of oligozoospermia mice,and then to explore Wuzi Yanzong Pills’s molecular mechanism of spermatogenesis is expected to provide a basic reference for the development and clinical application of Wuzi Yanzong Pills.Objective:To establish an animal model of oligozoospermia,to evaluate the effect of Wuzi Yanzong Pills on the animal model of oligozoospermia,and to further explore its molecular mechanism for the treatment of oligozoospermia.Methods:(1)Grouping and administration of experimental animals:divided into four groups:normal control group,oligozoospermia group,positive drug testosterone propionate group,and Wuzi Yanzong Pills treatment group,in which model group was given intraperitoneal injection of busulfan To establish a model of oligozoospermia,the positive drug control group was given testosterone propionate injection;(2)Determination of organ index of mouse testis and epididymis,analysis of HE-stained tissue sections of testis;(3)Use of fully automated mice The sperm analysis system evaluates and measures the sperm quantity and vitality of mice;(4)Evaluates the fertility of male mice by the number of litters of female mice after cage cages with female mice;(5)Obtains the testicles of mice by gene chip Gene expression profile,to explore the possible mechanism of action of Wuzi Yanzong Pills from the molecular level.Results:(1)The mice were injected intraperitoneally with a dose of 20 mg/kg of busulfan.The sperm count and activity were tested and the modeling was successful;(2)The testes organ index of mice in the model group decreased compared with the normal group.The testes organ index of mice in the Wuzi Yanzong Pills group approached the normal control group.HE staining results showed that the spermatogenic tubules in the model group were incomplete The proportion of spermatogonia decreased,and the ratio of spermatogenic tubules and germ cells in the Wuzi Yanzong Pills group was closer than that in the model group,and the number increased,reflecting the recovery effect of the drug;(3)Objectively evaluate the sperm quality of the mice in each group,and Wuzi Yanzong Pills shows the repairing effect on the sperm motility and quantity;(4)The number of litters in female rats of Wuzi Yanzong Pills is close to the normal group,and the number of litters in the model group and positive drug group is lower;(5)Through gene chip sequencing and bioinformatics mining,it is speculated that Wuzi Yanzong Pills mainly regulates the reduction of spermatogenic cells Splitting,testicular HR signaling pathway,testicular RAS signaling pathway,testicular cholesterol metabolism pathway,etc.affect spermatogenesis and testicular spermatogenesis,thus playing a therapeutic role in oligozoospermia.Conclusion:This study was the first to successfully establish an animal model of oligozoospermia,and systematically studied the spermatogenic effect of Wuzi Yanzong Pills,and evaluated the administration of normal mice,oligozoospermia mice,and Wuzi Yanzong Pills.The sperm motility parameters of the post-mice were tested on the fertility of the four groups of mice.The quality of the sperm was investigated from the source to the final result.Finally,the gene chip sequencing technology was applied to the genes of the mouse testis cells.The expression level was tested,and the molecular mechanism of Wuzi Yanzong Pills to improve sperm quality was discussed from the perspective of gene expression,which provided basic data for the further research of Wuzi Yanzong Pills.
