Abstract/Summary:
Nowadays many advances in science are made in search of discovering new technologies. Nevertheless, new technologies enable the further advancement of science. Hence, in order to further understand the world we live in, it is the merit of science to search for and push the limits never reached before and explore new possibilities. In such sprit, this thesis introduces a novel tool called nanoneedle. Nanoneedles were manufactured from assembly of carbon nanotubes into needle-like structure. Such structure itself, not only pushes the dimensional limits to molecular size diameters and aspect ratios to orders of magnitude greater than any structure with similar dimensions, but also enables the exploration of new limits in many applications some of which we have presented in the present document.;Nanoneedles can be used as novel electrochemical electrodes. The unique geometry and characteristics of nanoneedles enables them to improve the limits of the DNA concentration detection to 10 zeptomole. This is two or more orders of magnitude improvement from any previously reported detection based on electrochemical methods. Additionally, the simplicity of such detection setup based on nanoneedles makes it far more desirable than any sophisticated optical detection method. With their molecular dimensions and unique geometry, nanoneedles proved to be an ideal platform for single protein detection and manipulation that can push the limits of such studies to actual one or few immobilized and isolated proteins with zero background interference. Nanoneedles, with their dimensionally distinct structure, can be the only tools that can trans-feet sub-cellular components without killing the cell or altering its functionality, pushing the limits of transfection to single organelles within a cell. All these and many more foreseeable future applications and capabilities make nanoneedles a unique tool that can advance the boundaries of both science and technology to new horizons.
