With the rapid development of the global economy,more and more food varieties are accompanied by more and more food safety problems.From the collection and transportation of raw materials,to the processing and packaging of production workshops,and finally to the storage and sale of food after entering the market,there are a lot of potential food safety hazards in every link.One of the most unavoidable is the ubiquitous pathogens in the environment.Based on methods of metabonomics and genomic information,the differential metabolites of Staphylococcus aureus(ATCC6538),Salmonella(ATCC 14028)and Aspergillus flavus(CGMCC 3.6431)in culture medium,milk,spiced beef,dried bean curd and beef jerky were studied by gas chromatography-mass spectrometry(GC-MS)and headspace solid phase microextraction(HS/SPME).Genome information of three pathogenic bacteria was obtained.There were 80 different substances in Staphylococcus aureus,81 different substances in Salmonella and 33 different substances in Aspergillus flavus.Combining metabonomics with genomic information analysis,the following conclusions can be drawn:1.According to the optimized metabolomics research method for the purpose of this study,uncontaminated and contaminated food can be clearly distinguished,and the simpler the substrate components,the smaller the amount of samples required;otherwise,a large number of experiments are needed to clearly distinguish them.2.Through principal component analysis and orthogonal partial least squares discriminant analysis of experimental data of pathogens and blank groups,19,16 and14 potential characteristic differential metabolites were screened out in various substrate models of Staphylococcus aureus,Salmonella and Aspergillus flavus,respectively.3.Through the genomic analysis of each pathogen,the network diagram of the metabolic pathways of each pathogen were constructed.It was found that the higher the evolutionary level of organisms,the more abundant the types of metabolic pathways and the more advanced metabolic pathways.4.The differential metabolic pathways screened by the genomic information of each pathogen were analyzed,and the metabolic pathways of acetic acid,ethanol,glycerol and 1-butanol in three pathogenic strains were finally determined.
