ObjectivesIn this study,we investigated the effects of 2 hybrid scaffolds: Gelatin/polycaprolactone(PCL)and Collagen/PCL on proliferation and differentiation of bone marrow endothelial protenitor cells(BEPC).The same procedures were used to investigate the effects of 3D culturing on BEPC.We expect these will provide new ideas for tissue engineered corneal endothelium.Materials and Methods1?Gelatin/PCL and Collagen/PCL hybrid scaffolds were manufactured by electrostatic spinning and observed under the scanning electron microscope.Then BEPC were cultured on the 2 scaffolds and the cell morphology,adhesion,proliferation and stem cells characteristics were detected.2?BEPC were cultured on 3D dishes,and then the morphology,adhesion,proliferation and stem cells characteristics of BEPC were detected.Results1?BEPC had an expected growth tendency on Gelatin/PCL and Collagen/PCL.Gelatin/PCL and Collagen/PCL could promote the adhesion and proliferation of BEPC,the markers of differentiation and proliferation were also increased.2?BEPC on 3D dishes were floating tumor spheres.3D culturing could promote the proliferation of BEPC and markers of stem cells were also increased.Conclusions1?Gelatin/PCL and Collagen/PCL could promote the proliferation of BEPC and had a certain effects on differentiation.There were no significant differences between the 2 hybrid scaffolds in this study.2?3D culturing could not only promote the proliferation and differentiation of BEPC,but also facilitate the maintaining of stem cells characteristics of BEPC.3D culturing is an efficient culturing method and could be repeated.
