Objective: The damage and protection of low dose ionizing radiation are of great significance.To study the role of GSTP1 in low dose ionizing radiation damage.In-depth clarification of the biological effect of ionization radiation on cells,in order to further explore the application range of ionizing radiation in the medical field and provide theoretical foundation and experimental basis for radiation damage protection,treatment.Method: Peripheral blood specimens are collected healthy adults,people engaged in radiation,radiotherapy.QPCR detects the expression of GSTP1 in peripheral blood.Establish irradiation cell model,construct GSTP1 overexpresses cell line,Application of cell culture,flow cytometry,scanning electron microscopy,Transmission electron microscope,MTS to study the effects of ionizing radiation on the biological effects of cells.BSP technology detects the effect of ionizing radiation on the methylation of GSTP1.QPCR?Western Blot detect the expression of ionizing radiation on GSTP1.Western Blot detects the expression of NRF2,Caspase-3,JNK,and Bcl-2.Result: Compared with healthy adults,the expression of GSTP1 m RNA in the peripheral blood of radiation personnel decreased.Low dose ionizing radiation can cause cell morphological changes.Inhibit cell proliferation and block cell cycle in S phase,Promote apoptosis.Overexpression of GSTP1 can alleviate cell viability decline,cell cycle arrest and cell apoptosis induced by low dose radiation.Low dose ionizing radiation results in an increase in methylation of GSTP1 gene and can down-regulate the expression of GSTP1.Overexpression of GSTP1 can up-regulate the down-regulation of GSTP1 induced by low dose radiation.Low dose ionizing radiation can affect expression of GSTP1 related signal molecules.Conclusion: Low dose ionizing radiation has damage to cells,which can affect cellular biological effects.The overexpression of GSTP1 can alleviate the biological effects of low dose radiation on cells.GSTP1 is involved in the regulation of low-dose ionizing radiation damage and the expression of its related signaling molecules.
