Research findings in recent years,inhibition of p53 increased these adipogenic markers induced by BMP9.Inhibition of TGF-?1 attenuated the effect of p53 on enhancing BMP9-induced osteogenic markers in C3H10T1/2 cells.Inhibition of p53 decreased the ALP activity and protein level of OPN induced by BMP9,but it was almost reversed by TGF-?1over-expression in C3H10T1/2 cells.In the process of adipogenic differentiation,specific inhibitors of TGF-? 1 weakened the inhibitory effect of p53 on BMP9-induced adipogenic markers,p53 specific inhibitor enhanced the BMP9-induced adipogenic markers which were almost diminished by exogenous TGF-? 1.In addition,p53 promotes the expression of TGF-?1,and inhibiting p53 reduces the expression of TGF-?1.Our research results indicate that p53 is involved in the process of BMP9 regulating the osteogenic differentiation of mesenchymal stem cells,and the promotion mechanism of BMP9 osteogenic differentiation may be accomplished by regulating TGF-? 1 to further affect BMP9-regulated adipogenic differentiation.
