The formation and homeostasis of blood vessels play a vital role in animal life.The complete network of blood vessels in animal's body provides the necessary framework for blood flow.It is required for effective delivery of oxygen and nutrients and flushing out carbon dioxide and metabolic wastes.It is also important in delivery of blood cells,lymphocytes,hormones,and proteins to the various parts of the body.The formation and homeostasis of blood vessels is a complex process involving multiple signaling pathways.In recent years,more and more studies have proven that the BMP9/ALK1/Endoglin signaling pathway plays an important role in regulating angiogenesis and blood vessel homeostasis,and abnormal signaling can lead to hereditary hemorrhagic telangiectasia(HHT)and other diseases.In recent years,researches on BMP9/ALK1/Endoglin signal pathway have made many breakthroughs.However,little is known about the interaction between Endoglin and ALK1,and the molecular mechanism of Endoglin/ALK1 recognizing BMP9,I designed and constructed different eukaryotic expression vectors,compared different expression systems and purified the Endoglin/ALK1 heterodimer.Meanwhile,using the established Endoglin/ALK1 heterodimer expression system,I compared the binding ability of two BMP9 isoforms to Endoglin,ALK1 and the Endoglin/ALK1 heterodimer.These studies have provided a starting point for future studies of the structure of Endoglin/ALK1 heterodimer,and the role of BMP9/ALK1/Endoglin pathway in vascular remodeling and homeostasis.
