Objectives To screen the differentially expressed genes of coagulation in hepatocellular carcinoma and adjacent tissues by bioinformatics technology,so as to assist in early diagnosis and prognosis judgment of patients with hepatocellular carcinoma.Methods Differential analysis of 94 coagulation-related genes was performed with 50 pairs of HCC and adjacent normal tissues in The Cancer Genome Atlas(TCGA).381 TCGA samples with follow-up information(overall survival and relapse-free survival and survival status)and gene expression profiles were analyzed by univariate Kaplan-Meier survival analysis,and 358 TCGA samples with complete clinical information were analyzed by multivariate Cox regression.Patients were grouped according to their prognosis.Gene set enrichment analysis(Gene Set Enrichment Analysis,GSEA)was used to determine potential enrichment signaling pathways between high and low risk groups.Two independent HCC data sets were used to validate the results of the high and low risk groups.Immunohistochemical analysis was carried out to verify the expression of proteins related to the enrichment pathway in liver cancer tissues.The correlation between ITGA2,GP6 and CDH1,VIM,CCND1 gene expression was analyzed by R language.Results 1 Analysis of liver cancer and paracancerous samples showed 28 differential expression genes related to coagulation.2 Further survival analysis showed that ITGA2 and GP6 jointly affected the total progression-free survival of patients.3 GSEA results showed that Epithelial to Mesenchymal Transition(EMT)related pathways were significantly enriched in high-risk groups.EMT activated associated genes were upregulated in high-risk groups,and EMT inhibited associated genes were downregulated.4 EMT pathway-related proteins E-cadherin,Vimentin and cyclin D1 were significantly different in liver cancer and paracancerous tissues.5 ITGA2 was not correlated with the expression of CDH1,but positively correlated with the expression of Vim and CCND1.The expression of GP6 was positively correlated with the expression of CDH1,VIM and CCND1.Conclusions The expression of coagulation related genes ITGA2 and GP6 has an influence on the prognosis of liver cancer patients,which may be related to the activation of EMTrelated pathways.Figure10;Table3;Reference 152...
