Objective:The aim of our study was to investigate the prognostic significance of CDCA8(cell division cycle associated 8)and its possible mechanism of dysregulation in lung adenocarcinoma(LUAD)based on bioinformatics database,which provides a new therapeutic targets and prognostic markers for LUAD.Methods:CDCA8-related data,including gene expression,mi RNA expression,protein expression,gene copy number variation(CNA),DNA methylation data and clinical information,were obtained by Gene Expression Profile Analysis Interaction Analysis(GEPIA),Gene Expression Omnibus(GEO),the Cancer Genome Atlas(TCGA),Oncomine data base,UCSC Xena website,the Human Protein Atlas(THPA),c Bio Portal data base.Then CDCA8 expression level,CDCA8 protein expression level,regulatory mi RNA expression level of CDCA8 prediction and the clinical implication of CDCA8 were identified by t test and Chi-square test.The prognostic value of CDCA8 in LUAD were evaluated by Kaplan-Meier method,Cox regression analysis.GO,KEGG enrichment analysis and Gene set enrichment analysis(GSEA)was used to screen biological pathways correlated with CDCA8 expression in LUAD.Results:CDCA8 was significantly upregulated at gene levels and protein levels in LUAD tissue compared with normal lung tissues(P<0.05).Kaplan-Meier analyses suggested that CDCA8 gene expression was correlated with overall survival(OS)and disease-free survival(DFS)in LUAD.High CDCA8 expression was associated with more unfavorable overall survival(OS)and diseasefree survival(DFS)than low expression(all P<0.05).Cox risk proportional regression model analysis showed that overexpression of CDCA8 mRNA was an independent prognostic risk factor for overall survival(OS)and disease-free survival(DFS)(all P<0.05).The up-regulation of CDCA8 gene expression was positively correlated with the amplification of copy number variation(CNV),but negatively correlated with methylation(P<0.05).And hsa-mi R-140-3P downregulation were associated with CDCA8 upregulation in LUAD.GO,KEGG enrichment analysis and GSEA revealed that CDCA8 may be involved in cancerpromoting effect of LUAD mainly through cell cycle,DNA replication signaling pathway,spliceosome(all P<0.05).Conclusion:CDCA8 is up-regulated in lung adenocarcinoma,which is associated with CDCA8 DNA copy gain and has-mi R-140-3p downregulation.It may be the one of the independent prognostic risk factors.CDCA8 may be involved in cancer-promoting effect of LUAD mainly through cell cycle,DNA replication signaling pathway,spliceosome.
