Domestic garbage in Chinese cities is mainly transported to landfill for burial.This way of garbage disposal has caused a great impact on the surrounding soil,environment and people's production and life.Data show,that the leachate produced in the landfill process changes the nature of the surrounding soil through surface runoff,and the heavy metals and other substances accompanying the landfill not only harm the growth of crops in the field,but also pollute people's living environment and health with the harmful and unpleasant gases emitted from the garbage.There are also data,showing that landfills mainly change soil properties by affecting microorganisms in the soil.Next-generation sequencing is the most common type of gene sequencing at present.The main reasons for choosing this generation technology are as follows: first,it can detect hundreds of thousands or even millions of nucleic acid molecules at a time,saving the time needed for sequencing;Second,its testing cost is relatively affordable compared with the first generation,which is also one of the reasons why sequencing companies regard the Next-generation sequencing as the mainstream sequencing.In order to explore the impact of municipal solid waste(MSF)on microbial species,community composition and richness in the surrounding soil,the semi-living soil,near soil and remote control soil of a landfill site in Guiyang were collected for detection and analysis.In order to ensure the reliability of the experiment,each group of soil was evenly mixed and divided into three groups.A total of nine experimental data were collected,to analyze the changes of microbial diversity based on the secondgeneration Illumina Novaseq6000 sequencing platform.In this paper,Alpha diversity analysis was conducted on the experimental data,that met the requirements after sequencing,to measure the species richness of each sample and the diversity data of each value of the species,and the similarity of species diversity among each sample was compared with Beta diversity analysis.The results showed that a total of702176 pairs of reads were sequenced from 9 samples,and 698935 pairs of Clean reads were generated after double-end reads quality control and splicing.At least 63529 Clean reads were generated from each sample,with an average of 77659 Clean reads.According to shannon index,the microbial species diversity of the three groups of soil was rich.Semi-living soil > near soil > remote control soil.After analysis,the heavy metals in the garbage in the associated soil inhibit the diversity of microorganisms in the soil,while the long-term leachate in the near soil through surface runoff has an impact on the diversity of microbial species in the soil,which increases the diversity of microorganisms and changes the nature of the surrounding soil.Latescibacteria and Nitrospirae did not appear in the associated soil of landfill sites,while Rokubacteria had a low content in the associated soil.The effects of MSW in landfill on soil microorganisms were analyzed,and it was suggested that the dominant bacteria in MSW could be used to improve soil fertility and soil properties.In the later stage of the experiment,the potential functions of microorganisms in the soil were predicted,hoping to provide effective help for people's production and life by analyzing the dominant species and functions of microorganisms.The experimental results can also provide data reference and theoretical guidance for the impact of landfill leachate on the diversity of microorganisms in the soil.
