| Fuzhuan Tea is one of the unique black teas in our country,because it makes use of summer and fall tea resources.The products have the ability to invigorate the stomach,lower blood pressure and blood fat,which is enjoyed by people.Fuzhuan Tea can produce yellow cleistothecium in the process of fermentation,commonly known as the "golden flower"."golden flower" plays a crucial role in the process of Fuzhuan Tea quality,so the number of "golden flower" is taken as an important indicator to evaluate the quality of Fuzhuan Tea.Currently the detection of "golden flower" in the process of producing and starter cultures used in the process of promoting "golden flower",has always been great issues to Fuzhuan Tea’s producer and researchers,which is also a necessary condition on technology improvement of Fuzhuan Tea.In this study,On the one hand,by combining bioinformatics analysis and experimental verification method and strictly screening all sequenced Eurotium cristatum genomes for five times,specific orphan genes are obtained.Meanwhile designing its specific primers can be used to detect "golden flower" in the producing process of Fuzhuan Tea.At the same time we can find out the earliest technical process of "golden flower" bacteria phase;This study,on the other hand,through the matrix of black shai,adopting radiation sterilization,explore the single factor water content,sucrose,the best inoculation capacity.Then with the help of Design Expert,through comparing 17 groups of experiments finding out the best medium with the largest sporulation quantity,prepared the starter ZD1.At the same time,also selecting bran as matrix,through 121 ℃ high pressure steam sterilization for 20 mins,explore the single factor water content,Na Cl,the best inoculation capacity.Then with the helpof Design Expert,finding out the best medium with the largest sporulation quantity through comparing 17 groups of experiments,prepared the starter ZD2. |
