As a key species in the Southern Ocean’s bio-chain,Antarctic krill plays a key nutritional role in the Antarctic food chain.The physiological research on Antarctic krill is a hot research topic today.Anions and amino acids have the functions of regulating osmotic pressure,acid-base balance and regulating growth and reproduction during the growth of Antarctic krill.This study will determine the anion and three amino acid contents of Antarctic krill,and provide data support for the physiological process of Antarctic krill.And the laboratory culture of Antarctic krill is becoming more and more extensive.The value of this study can provide some theoretical support for the feed conditions in the Antarctic krill feeding process.There are two parts in this study.The first part is mainly to distinguish the age and gender of Antarctic krill.It is divided into eight samples and eight different anions in eight samples,including fluoride ion,bromide ion and sulfuric acid,are determined.There are eight anions of root,chloride,phosphate,nitrate,nitrite and iodide.The obtained data were used to analyze the role of each ion in the growth and development of Antarctic krill,and the possible interaction between ions was speculated.The second part is to determine the content of three positively charged amino acids of arginine,histidine and lysine in Antarctic krill in different periods and different genders,and analyze the role of three amino acids in the growth and development of Antarctic krill,the correlation between each amino acid and the possible correlation between the amino acid and each anion.Antarctic krill is a high nutritional food source.In addition to fishing according to regulations,large-scale cultivation in Antarctica and its similar environment will be the future direction of change.The data of this study also provide some data support for the feedstuff addition conditions of Antarctic krill in the future.The content of eight anions in Antarctic krill was determined by ion chromatography.The analysis data showed that the chloride ion content was the highest among the eight anions,and it was 8449.47 μg/g at the fourth stage.At the same time,anions fluctuate obviously in the third and fourth phases.The contents of ions generally decreased in the second stage,increased significantly after the third stage,and gradually decreased in the fifth stage.The content of three amino acids in Antarctic krill was determined by TLC.The optimum extraction method of amino acids and the development system of the three amino acids were determined.Ultrasound extraction of amino acids was carried out by adding 40 m L absolute ethanol and 10 m L deionized water.The deployment system of arginine is n-butanol:formic acid: water = 15:3:2(V:V:V),histidine is n-butanol: anhydrous ethanol:concentrated ammonia water: water = 8:8:1:3(V:V:V:V),lysine is deployed by glacial acetic acid: water = 8:2(V:V).The analysis data showed that the arginine content was the highest in the third male stage,which was 20433 μg/g.Histidine content was the highest at the fourth stage,6240 μg/g.Lysine content was the highest at the fourth stage(23537.5 μg/g).The trend of histidine and lysine is similar to that of most anions,which first decreases and then increases.Arginine increased first and then decreased gradually. |
