| Oplopana elatus Nakai is a perennial shrub plant of the family Araliaceae and stichopus.Its medicinal properties and chemical composition are similar to ginseng,so it is known as "woody ginseng".According to the monographs of national medicine,it has the functions of nourishing yang,tonifying the collaterals,relieving cough,regulating Qi,removing blood stasis,etc.In recent studies,it has been found that the northeast ginseng can regulate immunity,improve metabolism,regulate blood pressure and blood sugar,suppress cancer Cough for a long time,external evil attack meridian and other aspects also have good therapeutic effects,especially in the anti-cancer area.The ginseng ginseng contains a variety of chemical components,the main active ingredient is the ginseng ginseng saponin,which belongs to pentacyclic triterpenoid saponins.It has a high content in its roots and rhizomes,which has great medicinal value and broad application prospects.Due to the lack of wild resources,the artificial cultivation technology is not mature,and the low content of adventitious root saponins of the ginseng ginseng cultivated in the bioreactor,how to increase the content of saponins in the northeast ginseng ginseng and enhance its biological activity has become a research hotspot.In recent years,studies on plant endophytes have shown that endophytes are involved in the conversion and synthesis of plant secondary metabolites,and can also independently produce secondary metabolites,which is an inexhaustible source for discovering new drugs and lead compounds.Therefore,in this study,endophyte strains were isolated from the ginseng ginseng and the biotransformation of endophytic bacteria was used to increase the total saponin content of the active ingredient in the roots of the ginseng ginseng.In this study,methods such as flat plate culture and four-zone line drawing were used to isolate and purify the endophytic bacteria of the ginseng ginseng from the northeast,and the endophytic bacteria were used for liquid fermentation of the ginseng ginseng to obtain the fermentation product of the ginseng ginseng.The method was used to determine the content of total saponins,to explore the influence of microbial strain inoculation amount,fermentation temperature,and fermentation pH on fermentation products,and to optimize fermentation conditions.We isolated 78 species of endophytic bacteria from the ginseng of Northeast China and selected three highly active strains,namely strains PDA7,NA22 and NA23.The fermentation experiment results showed that the content of ginsenoside reached the highest value of 54.597 μg/mL after 1 day of fermentation of strain PDA7;the content of ginsenoside reached 55.339μg/mL after 11 days of fermentation of strain NA22.After 11 days of fermentation with strain NA23,the content of ginsenoside reached 56.347μg/mL.The most favorable conditions for the liquid fermentation process of ginseng ginseng are:fermentation temperature is 30℃,initial fermentation pH is 7.0,and endophyte inoculation amount is 10%.Finally,the 16S rDNA sequence analysis method was used to identify the highly active endophytic strains.The results showed that strain PDA7 was Bacillus subtilis;strain NA22 was Agrococcus citreus;strain NA23 was Bacillus pumilus. |
