| Diabetes is a metabolic disease characterized by chronic hyperglycemia,which is closely related to factors such as genetics,living habits,work pressure,and diet.Diabetes is showing a younger trend in recent years,seriously endangering people’s health.Protein tyrosine phosphatase 1B(PTP1B)is a member of the protein tyrosine phosphatases(PTPs)family and a key negative regulator in the insulin signal transduction pathway.Therefore,PTP1 B is regarded as a target for increasing insulin sensitivity and treating diabetes.At present,although the traditional treatment of diabetes is effective,but it has some side effects.Therefore,there is an urgent need safer and more effective natural plant or food adjuvant therapy.According to the preliminary research of our group,we used water extract powder of7 kinds of food or medicinal and edible plant,including kelp,pumpkin seeds,and yam,A,B,C,D,supplemented with xylitol,to prepare a solid beverage(named “Kunnansu”auxiliary hypoglycemic solid beverage)that can assist in lowering blood sugar.The experiments proved that the Kunnansu can effectively inhibit the activity of PTP1 B,and has good specificity in vitro.Using mouse 3T3-L1 cells,we tested the effect of Kunnansu on the insulin signaling pathway at the cellular level.The results showed that Kunnansu can increase the tyrosine phosphorylation of cellular proteins and activate upstream and downstream molecules IRβ,ERK1/2 and GLUT4 in insulin signaling pathway,and Kunnansu can increase insulin sensitivity.This effect is probably due to the inhibition of PTP1 B by Kunnansu.In order to further study the hypoglycemic effect of Kunnansu in vivo,we constructed a mouse model of hyperglycemia with low insulin secretion.The diabetic mice were fed with Kunnansu by gavage every day for 31 days.Compared with the control group,Kunnansu medium-and high-dose groups can significantly reduce the fasting blood glucose level of Diabetic mice and increase the tolerance of model mice to glucose.In summary,we used the water extract powder of 7 kinds of food or medicinal and edible plant,supplemented with xylitol,to prepare a solid beverage named Kunnansu.Kunnansu had an insulin-like effect and could increase insulin sensitivity,and to a certain extent improved the symptoms of diabetic mice.It had the potential to become an auxiliary hypoglycemic health food.This research provided new ideas for the development of antidiabetic natural plant-based auxiliary hypoglycemic health food. |
