This paper studied the extraction of polysaccharide from Dictyophora indusiata and its effect on the growth and proliferation of Lactobacillus acidophilus and Clostridium butyricum.Polysaccharides have many functions such as anti-oxidation,anti-inflammation,anti-tumor,lowering blood lipid,and enhancing immunity,and have good nutritional and health value.However,the probiotic activity of DIP.indusiata polysaccharides was rarely reported.This paper intends to study the probiotic activity of DIP.indusiata polysaccharides.DIP was extracted by water bath extraction with ultrasonic-microwave assisted pretreatment,and separated and purified by fractional ethanol precipitation with different concentrations.The structure of DIP and its effect on the growth and proliferation of probiotics were studied,and the components of DIP were analyzed.The main research results are as follows:1.Extraction of polysaccharide from Dictyophora indusiata:The extraction of DIP was carried out by single factor experiments(ultrasonic power,microwave power,particle size,water bath temperature and solid-liquid ratio).Three factors and three levels that had great influence on the extraction of DIP were selected for orthogonal experiments.The optimal extraction conditions were as follows : ultrasonic power500 W,microwave power 200 W,particle size 80 mesh,water bath temperature100℃,solid-liquid ratio 1:70 g/m L,and the extraction rate of crude polysaccharide was 27.15%.2.Purification and analysis of Dictyophora polysaccharide : DIP was separated and purified by stepwise ethanol precipitation with different concentrations.Structure and composition analysis of DIP The structure and composition of DIP were analyzed by FTIR and Ultra-High Liquid UPLC-Q-Orbiap.The DIP contained β-glycosidic bond structure by FTIR analysis.The extract mainly contained 111 components,including 7polysaccharides,6 amino acids,1 polyphenol,3 organic acids,2 alkaloids,10 flavonoids,14 fatty acids and 3 vitamins.3.Effects of Dictyophora polysaccharide on the growth and proliferation of L.acidophilus and C.butyricum:DIP promoted the growth and proliferation of probiotics.L.acidophilus and C.butyricum were cultured on MRS medium containing different components of DIP,and the 24 h growth curves of the two strains were plotted.The results showed that in vitro simulated artificial gastric juice growth curve had the best effect on bacteria protection and proliferation,and DIP was higher than that of inulin and sugar-free blank group.In the growth curve of DIP on L.acidophilus.acidophilus and triple C.butyricum 6 h in vitro simulated artificial intestinal fluid,DIP had the best growth promotion effect in artificial intestinal fluid,and the growth promotion effect of DIP group was higher than that of inulin and blank group.In the growth curves of L.acidophilus and C.butyricum for 6 h,the DIP group had the best protection effect than inulin and blank control group.When polysaccharides with different components and different concentrations were used as carbon sources for culture,the best polysaccharide for L.acidophilus culture was DIP with 0.3% content,and only 0.2% and 0.3% DIP were higher than those of inulin.Different DIP1 and DIP2 were slightly lower than those of the control.0.1% DIP could replace 0.2%inulin for the proliferation of C.butyricum.0.3% DIP had the best promotion effect,and DIP1 and DIP2 at different concentrations were lower than those of inulin.In summary,crude polysaccharides from Dictyophora indusiata had obvious growth and proliferation effects on L.acidophilus and C.butyricum,and had probiotic activities.
