This paper explores the qualitative and quantitative methods of gas chromatography combined with negative chemical ion source mass spectrometry(GC-NCI-MS)for fatty acids,organic acids and amino acids,including: pretreatment of fatty acids,organic acids,amino acid derivation;The quantitative analysis results of GC-NCI-MS method are evaluated and applied in practice.The GC-NCI-MS method and gas chromatography combined with electron bombardment ion source mass spectrometry(GC-EI-MS)compared the detection of fatty acids,organic acids,amino acids,and the results were as follows:First,this study compared chloroform(Chloromethal MCF,Ethyl chlorate ECF,Propyl chloroformate PCF)and sulfuric acid-methanol solution derived from fatty acids,organic acids,amino acids,chloroformate derived best conditions are: water bath temperature of 80 degrees C,water bath time 60 min,dichloromethane Extraction,the largest number of derivatives,up to 8 kinds;The GC-NCI-MS method was tested for methodological indicators,including linear index,minimum detection limit(LOD),minimum quantitative limit(LOQ).),repeatability,reproducible,accuracy(in terms of recovery rate),degree(variation coefficient CV),the method is shown as linear good,wide range of application,high sensitivity,high accuracy and degree;Finally,in the detection of the actual samples(fatty acids of fermented milk,organic acids and amino acids of mushrooms),this study compares and summarizes the advantages and disadvantages of chloroformate derivative combination GC-NCI-MS method and conventional saponification derivation combination GC-EI-MS method.The chloroformate derivative combination GC-NCI-MS method can meet the fatty acids,organic acids,amino acids at the same time qualitative and quantitative,compared with conventional saponification derived combination GC-EI-MS method,chloroform can be derived at room temperature,and medium,short-chain fatty acids,polyxyl/hydroxy organic acids,branched chain amino acids and so on have better derivative effects.More importantly,GC-NCI-MS method can retain quasi-molecular ion information,strong anti-substitial interference ability,good selectivity,simple mass spectrometography,easy manual analysis and so on.The method of detecting fatty acids,organic acids and amino acids in combination with GC-NCI-MS will be of great application value in chemical,biological,medical and food processing active ingredient detection.
