| In the experiment,the recovery rate of precipitant,the concentration of precipitant,the extraction time and the temperature of sample were taken as the optimization items of sample pretreatment.After the pretreatment conditions were determined,the chromatographic column,the flow phase and the tapered voltage were taken as the optimized terms of the instrument conditions.The method has been verified.After the determination of the method,the machine was tested.The difference between the liquid phase method and the liquid phase method of gb tanesulfonyl chloride was analyzed.Different infant formula milk powders were tested(n=6),The results of taurine were compared in different laboratories by this method,and the difference was analyzed.Through experiments,it was found that 75% ethanol was the most suitable sample for precipitation.Ultrasonic extraction at 40 ℃ for 10 min was the best.The amide column was used in the machine condition.In mobile phase,0.1% formic-acetonitrile,gradient elution,mass spectrometry with cone voltage of 15 V and anion mode were selected.The best effect was obtained with an injection volume of 1 L.The linear relationship of taurine was good in the range of 0.04 ~ 10.0 g/m L.The detection limit of the instrument was 6.5ng/m L and the quantitative limit was 21.6 ng/m L.Instrument precision(parallelism)was 1.4 %,sample precision(repeatability)was 4.7 %,The recovery of external standard method was between 99.6% ~ 101 %.Taurine in infant formula was detected by this method(n=6).There was no significant change in the detection results compared with the national standard liquid chromatography(P > 0.05).Different infant formula milk powders were tested(n=6),Compared with the taurine measured in different laboratories by this method,the two results were P > 0.05.UPLC-MS method meets the requirements of detection.Compared with the liquid phase method,the recovery was higher.Compared with danesulfonyl chloride derivation,it can save nearly 2 hours and improve the efficiency.There was no significant difference between the two laboratory tests.Taurine in other infant formula milk powder was detected,and the results showed no significant difference from the labeled values.The results show that the method can be used to detect taurine in infant formula milk powder. |
