This topic effectively combines extrusion technology with modern extraction and separation technology of phytochemicals,and establishes an innovative method that can increase the extraction rate of effective components in Hs Pc-HA nutrient solution.Used in the treatment of malignant tumors and nutritional support in the later stage of treatment.Discuss its effects from both in vivo and in vitro,and clarify its mechanism of action at the molecular level.The main research contents are as follows:(1)Extrusion pretreatment was used to extract effective components,and Hericium erinaceus was taken as an example to determine the optimal extrusion expansion parameters and the effect of extrusion pretreatment on the extraction rate of polysaccharides.The optimized results of response surface analysis showed that the optimal extraction conditions of Hericium erinaceus polysaccharides were as follows:screw speed 400r/min,moisture content 16%,and extrusion temperature 120℃.After verification,it is determined that the extraction rate can reach up to 3.63±0.02% under these conditions.The effective components are matched according to the proliferation effect of splenic lymphocytes,and finally the four effective components of Hericium erinaceus polysaccharide: Acanthopanax senticosus polysaccharide: Poria cocos polysaccharide: Atractylodes lactone are configured in a ratio of 1:1:2:1 Hs Pc-HA nutrient solution.(2)Construct an IEC-6 cell injury model in vitro,and explore the effect of Hs PcHA at the cellular level.The MTT results showed that when 1.5 mg/m L cyclophosphamide(CTX)was treated for 48 hours,the IEC-6 cell injury model could be successfully constructed.And the best repair rate of Hs Pc-HA for IEC-6 cells can reach 47.18±1.23%.The results of cell scratch test and HE staining showed that the healing rate of the model group was 8.20±1.03% after the model was administered and cultured for 24 hours,and the healing rate of the Hs Pc-HA group could reach16.35±1.57%.It shows that Hs Pc-HA can promote IEC-6 cell damage migration,can improve the healing rate of cell damage,and can appropriately improve the cell survival rate.(3)Subcutaneous inoculation of S180 ascites tumor cells can successfully construct a tumor-bearing mouse model.After intraperitoneal injection of 12 mg/m L cyclophosphamide(CTX)for a week,a tumor-bearing mouse model after chemotherapy can be successfully constructed.Compared with the model group(CTX group),the body weight,food intake,and survival rate of mice were significantly improved after Hs Pc-HA was administered.The survival rate of the CTX group was only 62.5%.After the administration of nutritional support,the survival rate can reach75%,indicating that Hs Pc-HA can increase the survival rate of immunocompromised tumor-bearing mice,and prolong the survival time of mice with radiotherapy and chemotherapy.In addition,Hs Pc-HA can effectively promote the gastric emptying rate and small intestine advancing rate of tumor-bearing mice after chemotherapy,increase the chest and spleen index of mice,and reduce the ear swelling rate and granuloma inhibition rate of mice after chemotherapy.Certainly enhance immunity and antiinflammatory effects.The results of HE staining showed that Hs Pc-HA can reduce the damage of various tissues and organs caused by CTX.The results of biochemical and ELISA methods show that Hs Pc-HA can significantly reduce aspartate aminotransferase(AST),alanine aminotransferase(ALT),total bilirubin(TBIL),and urea in serum of tumor-bearing mice after chemotherapy Nitrogen(BUN),and at the same time can increase the serum immune factors superoxide dismutase(SOD),malondialdehyde(MDA),tumor necrosis factor-ɑ(TNF-α),interleukin-2(IL-2),The content of interleukin-8(IL-8),immunoglobulin M(Ig M)and immunoglobulin A(Ig A)can reduce liver and kidney damage and regulate the body’s immune function.Western Blot method was used to determine the protein content of nuclear transcription factor-кB(NF-кB p65)in the spleen tissue of mice,and to explain its anti-inflammatory mechanism.The results show that Hs Pc-HA can down-regulate the protein expression of NF-κB P65 in the spleen tissue of tumor-bearing mice after chemotherapy,and reduce the inflammatory response caused by CTX by inhibiting the conduction of NF-κB signaling pathway.
