| Salvia Miltiorrhiza Bunge has the functions of promote blood circulation and remove blood stasis,inhibite bacteria and inflammation,and anti-tumor.In this study,the volatile oil of S.miltiorrhiza was extracted from the root of S.miltiorrhiza by steam distillation method.The components of the volatile oil of S.miltiorrhiza,the antioxidative activity and tyrosinase inhibitory activity of volatile oil and ethanol extract from S.miltiorrhiza were studied:(1)Based on single factor and response surface optimization method,the optimum extraction parameters of the volatile oil from S.miltiorrhiza were obtained as followed:S.miltiorrhiza was about 1cm2,soak time was 3 h,extraction time was 8 h,addition of sodium chloride was 3.6%and ratio of material to liquid was 1:10.Under the optimum conditions,the actual extraction recovery of the volatile oil of S.miltiorrhiza is 0.032%.The optimization result showed that the addition content of sodium chloride was the principal factor affecting the extraction efficiency of the volatile oil from S.miltiorrhiza,followed by extraction time and ratio of material to solvent.The constituents of the volatile oil from S.miltiorrhiza were analyzed by GC-MS,25 constituents were identified.The major constituents are palmitic acid,linoleic acid and ferruginol(28.80%,26.81%,19.46%respectively),accounting for 75.07%of the total amount of the volatile oil.(2)The results related to volatile oil and ethanol extract from S.miltiorrhiza in certain reaction buffers indicated their effective antioxidant activity.The volatile oil of S.miltiorrhiza and ethanol extract of S.miltiorrhiza had effective scavenging effect on DPPH and ABTS free radicals systems.The scavenging effect of volatile oil of S.miltiorrhiza on hydroxyl radical was stronger than that of positive control VC,and the total reduction ability of volatile oil was better than that of ethanol extract.The results of antioxidant activity related to complex of dandelion extract and volatile oil of S.miltiorrhiza showed that the DPPH free radical scavenging effect of the complex was obviously enhanced,the scavenging effect was similar to that of the positive control VC,the synergistic coefficient was SE>1,which showed the synergistic antioxidant effect.The scavenging effect of volatile oil from S.miltiorrhiza on hydroxyl radical was enhanced by the addition of dandelion extract.(3)The results showed that ethanol extracts of S.miltiorrhiza and the volatile oil of S.miltiorrhiza have good inhibitory effect on tyrosinase activity,the inhibition of three substances to tyrosinase was stronger than that of ethanol extract of S.miltiorrhiza(IC50=0.41 mg/m L)>volatile oil of S.miltiorrhiza(IC50=0.96 mg/m L)>Arbutin(IC50=1.90mg/m L).The inhibitory kinetics of tyrosinase showed that ethanol extract of S.miltiorrhiza inhibited tyrosinase by both reversible competitive and non-competitive mixed type inhibition,the type of tyrosinase inhibition by the volatile oil from S.miltiorrhiza was reversible competitive inhibition.The inhibition of tyrosinase by the dandelion extract was non-competitive and uncompetitive mixed type II,and there was no significant change in tyrosinase inhibition rate before and after mixed the volatile oil of S.miltiorrhiza and dandelion extract,the results showed that there was no antagonism among the the volatile oil of S.miltiorrhiza and dandelion extract,and no adverse effect on the inhibition of tyrosinase activity.(4)The volatile oil of S.miltiorrhiza was applied in cream and handmade soaps,the results showed that its physical and chemical properties and sensory evaluation were adapted for the current industry standards,so the volatile oil of S.miltiorrhiza could be used as a whitening and anti-oxidation substance in cream and other daily chemical products in future. |
