Exercise-induced fatigue is a state that the body cannot maintain at a specific level after a series of exercises,accompanied by changes in physiological indicators.Foodborne active ingredients are widely sourced and safe,and have broad development prospects.The role of foodborne active ingredients in alleviating exercity-induced fatigue has attracted much attention.This paper selected five typical foodborne active ingredients(tea polyphenols,ginseng polysaccharide,Lycium ruthenicum polysaccharide,ginseng saponin,sea cucumber peptide),by constructing the animal model,judge its anti-fatigue effect,analyze the micro RNA expression changes,to explore the molecular mechanism of foodborne active ingredient fatigue resistance,the main contents are as follows:(1)To explore five kinds of active ingredients of exercise-induced fatigue in mice and the effect of exercise fatigue in mice model,this paper on the mice were randomized,10 in each group,each type of foodborne active ingredient is set low,medium and high three doses of concentration,meanwhile set up blank lavage distilled water group,model group,compound group as positive control group lavage compound danshen dropping pill.The mice were given intragastric gavage once a day for consecutive 28 days.Swimming was performed half an hour after the end of intragastric gavage.Swimming exhaustion experiment was performed half an hour after the end of intragastric gavage on the last day,weight-bearing(5%of body weight)was applied to the tail of the mice,and the time was recorded.The experimental results show that,after multiple physiological factor analysis,tea polyphenols,ginseng polysaccharides can reduce the concentration of lactic acid,increase the concentration of lactate dehydrogenase,reduce the concentration of urea nitrogen,increased superoxide dismutase concentration,reduce the concentration of malondialdehyde,anti-fatigue effect is good,Lycium ruthenicum polysaccharide,ginseng saponin and sea cucumber peptide to some extent have a relief on exercise-induced fatigue,But the dose still needs to be improved.(2)Through previous studies,it was proved that tea polyphenols and ginseng polysaccharide had better anti-fatigue effect.Further high-throughput micro RNA sequencing analysis was conducted on the serum of these two groups of mice,and it was found that there were 23 differentially expressed micro RNAs in tea polyphenols group and3 differentially expressed micro RNAs in ginseng polysaccharide group.mi R-143-3p and mi R-200c-3p were differentially expressed micro RNAs shared by the two groups,and the anti-exercise fatigue effect of tea polyphenols and ginseng polysaccharide might be regulated by these two targets.(3)The main component of tea polyphenols is epigallocatechin gallate(EGCG).In order to further explore the molecular mechanism of exercise-induced fatigue,cardiomyocytes related to exercise fatigue were selected for study,which can cause injury and apoptosis of cardiomyocytes during exercise.In this study,myocardial cell apoptosis model was induced by H2O2for in vitro culture,and the expression levels of cupramine oxidase 1(AOC1)m RNA and mi R-16-5p were detected.Our study found that EGCG can inhibit the up-regulation of mi R-16-5p expression,and then target the expression of AOC1and inhibit the apoptosis of cardiomyocytes,thus alleviating the state of exercise fatigue,which also preliminarily confirmed the regulatory role of micro RNA in exercise fatigue.
