| Omeprazole is now widely used in clinical digestive ulcer disease,since it has high efficiency in acid suppression;but it’s unstable in acid susceptible,so the formulations are mostly enteric formulations;however enteric formulation can’t alleviate the suffering of patients at time,because it reduces the initial acid-suppressing effect of omeprazole.So in this paper we prepared immediate-release omeprazole pellets by selecting the types and the amount of antacid,and analyze it though vitro dissolution.Prepared immediate-release omeprazole pellets to improve the stability of it in acidic environments,and analyze it though vitro dissolution.The formulation and process parameters were optimized by single factor design.To improve the stability of omeprazole in acidic conditions,an antacid,sodium bicarbonate,was adding into the formulation to increase the microenvironmental pH.The optimal pellets were prepared with the blank pores of microcrystalline cellulose,1.5%hypromellose solution as a adhesive,and amounts of sodium bicarbonate and microcrystalline cellulose.The final release effect and stability of immediaterelease omeprazole pellets was valued through vitro dissolution.The dissolution of immediaterelease omeprazole pellets at 15 min in pH 1.2 hydrochloric acid was above 85%,and the product could maintain the stability of omeprazole in 1 h. |
