Ficus tikoua Bur,a creeping woody liana shrub of the genus Ficus,and its root、stem、fruit all can be used as medicine,has been widely used in the minority areas which located in the southwest of China.To make better use of this wild resource,the antifungal activity of the methanol extract of the aerial parts of Ficus tikoua were studied systematically in this paper.To provide a reliable theoretical basis for the direct development and utilization of this plant,the standard HPLC profiles of the eight tested flavones(D1-D8)were established based on the previous research to investigate.The results reveal the differences in the contents of antifungal active ingredients from different areas.In this paper,the antifungal activity of the crude extracts from the aerial parts of F.tikoua against Pseudoperonospora cubensis and Phytophthora infestans were evaluated.The inhibition rates on the release of the zoospore of P.cubensis and P.infestans of the extract,and the inhibition rate on the mycelium growth of P.infestans,were tested by spore germination and mycelial growth methods,respectively.The results of spore germination tests showed that the crude extracts had good inhibitory effect on the two tested pathogens,the inhibition rates were more than 90% when the concentration was 1000μg/ml,and the inhibition rates also still could reach to above 75%.The results of mycelial growth test revealed that the mycelial growth of P.infestans could be effectively inhibited at the concentration was 1000μg/ml until 168 h and 240 h,and the inhibition rate were 96.97% and91.67%,respectively.The inhibition rate could reach to above 65% after 96h~240h when treated at the concentration of 500μg/ml.The pot experiments showed that the control effects of 20% EC of F.tikoua(500-fold dilution)on the cucumber downy mildew was above 85%,and which is comparable to the commercial fungicide dimethomorph(2000-fold dilution).The botanical formulation also showed good control effects on potato late blight,and its protective efficacy was 93.82%.Field efficacy trials showed that the control effects of the botanical formulation on cucumber downy mildew was comparable to dimethomorph,and the average control effects was above 75%.To investigate the QC for the botanical fungicide and to provide a comprehensive and reliable theoretical basis for the development and utilization of this plant,a HPLC profiles of flavones,D1-D8,were established based on the method of HPLC-PDA-ESI/MS.The contents of flavones in the aerial parts of F.tikoua from Kaiyang of Guizhou,Hongya of Sichuan,Hejiang of Sichuan,Kunming of Yunnan,Ankang of Shanxi and Xishui of Guizhou,were determined by using the established method.The results showed that all of the flavones could be detected in different samples,but there were high difference in the content of eight active compound,especially for D2 and D6.The in vitro an in vivo antifungal activity and the multi-components analysis of F.tikoua were investigated in this paper.The results will provide theoretical guidance for the development of this botanical fungicides.
