Research On Food Quality And Safety And Drug Analysis Using Chemical Multi-way Calibration Method

The calibration in chemometrics is often applied to quantitative analysis.The data obtained in modern analytical chemistry has changed from single point data to multi-way data and people’s food,clothing,shelter and transportation involve many things.In recent decades,the quantitative analysis of related substances has attracted more and more attention,and chemometrics have successfully solved this problem with multi-way calibration,and have been successfully applied to the environment,food,medicine and other complex systems.This paper makes full use of the advantages of chemical multi-way calibration to solve quantitative and related problems,and further obtains useful information,and carries out a series of applied researches in the field of food,medicine and environment related to human health,mainly including the following four aspects:In Chapter 2:In this chapter,a new method for the determination of bisphenol A(BPA)and diphenyl carbonate(DPC)in polycarbonate plastic water bottles was presented by three-dimensional fluorescence and chemometrics.This method only needs some simple dissolution and a little solvent.The idea of green chemistry is advocated,and the detection is quick and sensitive.The method proposed in this chapter allows the existence of unknown background interference in the actual sample and allows accurate quantification of three kinds of plastic samples with spectral overlap,which benefits from the“second-order advantage”of multi-way correction method in chemometrics.The limits of detection of the two analytes were 0.040 ng m L-1and 1.180×103ng m L-1,and the limits of quantitative were 0.110 ng m L-1and3.580×103ng m L-1,respectively.In addition,the average recovery for BPA and DPC were 99.4%and 83.5%,respectively,and the relative standard deviation were less than 2.3%.The results show that this method is effective and sensitive for the determination of BPA and DPC in different polycarbonate plastic water bottles.It only needs simple sample pretreatments and costs less.In Chapter 3:This chapter developed a method based on high performance liquid chromatogram-diode array(HPLC-DAD)for the simple and rapid simultaneous determination of plant growth regulators(PGRs)in lemon,peach and plum.The plant growth regulators measured in this chapter included abscisic acid(ABA),3-indolebutyric acid(IBA)and 6-benzyl aminopurine(6-BA),and their elution time were 2.65 min,2.83 min and 3.22 min,respectively.The separation was carried out by gradient elution.The column temperature was 40℃and the flow rate was 1 m L min-1.The linearly dependent coefficient of 6-BA,ABA and IBA in this method are good(99.6%,99.9%and 99.8%),and the the limits of detection of 6-BA,ABA and IBA are0.010μg m L-1,0.010μg m L-1and 0.040μg m L-1,respectively.Their standard recovery were kept in the range of 92.4-112.6%.The repeatability and reproducibility of the method were evaluated by intra-day and inter-day precision analysis.The method in this chapter has the advantages of simple fruit pretreatments and fast experimental speed,and can simultaneously detect multiple PGRs under the interference of other fruit components.In Chapter 4:In this chapter,the contents of the psychotropic drugs donepezil hydrochloride(DON)and trazodone hydrochloride(TRA)in human body fluids were determined by using the ANWE algorithm.Although DON,TRA and plasma background interference have significant peak overlap,pure DON and TRA signals in plasma can be obtained and reliable qualitative and quantitative results can be obtained.The standard recovery of DON and TRA were 104.1%and 86.5%,and their standard deviation were lower than 4.3%and 2.2%,respectively.In addition,the accuracy of the method was assessed using quality factors such as sensitivity,selectivity,detection limits,and quantification limits,as well as intra-day and inter-day precision.There is no significant difference among ANWE,ATLD and SWATLD algorithms.The results are satisfactory,indicating that the proposed method can be used for the accurate,rapid and non-interference determination of DON and TRA in the body fluids of psychiatric patients.In Chapter 5:In this chapter,three-way fluorescence and the two-order calibration method are proposed for the determination of bambuterol in human body fluids.Despite using the less interference separation of body fluids and having the significant spectral overlap between bambuterol and the background,the proposed method could accurately quantify bambuterol in plasma and urine samples.The standard recovery of bambuterol was 100.1%and the standard deviation was lower than 6.3%.The calculated results include high sensitivity,good selectivity,low detection limit and quantification limit,and high intra-day and inter-day precision.Therefore,the proposed method is expected to be used for real-time quantification of bambuterol in the body fluids of patients with respiratory diseases.
