Myrica rubra(Lour.)S.et Zucc.is native to China.As one of the famous fruits in in regions south of the Yangtze River,its fruit is delicate and juicy with sweet and sour taste,which is popular among consumers.In addition,Myrica rubra are easy to cultivate,with long economic life and low planting cost.By the end of 2016,the total area of Myrica rubra cultivation in our country had reached 333,300 hectares.Because of abundant anthocyanins,Myrica rubra has high edible and medicinal value.However,due to the instability of anthocyanin,its bioavailability is greatly affected.In this study,cyanidin-3-O-glucoside chloride(C3G)was taken as the research object.By using liposome encapsulation technique,the absorption effect of C3G nanoliposomes in gastric epithelial cells(GES-1 cells)as well as the antioxidant effect of C3G nanoliposomes on GES-1 cells was explored.1.Anthocyanin nanoliposomes were successfully prepared by ethanol injection.The particle size and encapsulation efficiency were 186.6±3.47 nm and 75.7%±1.01%,respectively.The polydispersity index(PDI)was 0.174±0.024.The storage stability and structural micro-evaluation were evaluated.It was found that the p H value of anthocyanin nanoliposomes remained stable and the particle size increased slightly in two weeks.2.The effects of anthocyanin nanoliposomes on gastric cells were evaluated by cell activity and apoptosis experiments and the mitochondrial microstructure.It was found that there was no significant change in mitochondrial structure,and anthocyanin nanoliposomes could prevent GES-1 cells from apoptosis.In the study on the absorption of anthocyanin nanoliposomes in gastric cells,it was found that gastric cells had a certain dose effect on the absorption of anthocyanin nanoliposomes(0-6h),which could be absorbed rapidly in a short time,and the average fluorescence value reached the maximum at 4h,and green fluorescence could be observed in the nucleus under fluorescence microscope.3.In vitro antioxidant experiments showed that anthocyanin nanoliposomes could effectively scavenge free radicals.After the GES-1 cells were induced by H2O2,the total antioxidant capacity(T-AOC)was significantly increased,and the content of malondialdehyde(MDA)was effectively reduced by adding anthocyanin nanoliposomes,indicating that C3G liposome can effectively enhance the antioxidant capacity of GES-1 cells.
