This paper mainly studied the secondary metabolites and their bioactivities of two endophytic fungi Penicillium crustosum JT-8 and Penicillium expansum JT-10,which isolated from tubers of Paris polyphylla Smith var.yunnanensis.The first chapter summarized the biological activity of the endophytic fungi and the secondary metabolites of some genus from Paris polyphylla Smith var.yunnanensis.In the second chapter, the solid fermentation product of P.crustosum JT-8 was systematically isolated and analyzed.Fifteen compounds were obtained and identified as: one new cycloheptane derivative: crustane(1);the others: quindoline(2),viridicatin(3),3-indolethanol(4),roquefortine C(5),(2S)-hydroxyroquefortine C(6),ergosterol(7),β-sitosterol(8),1,1’-binaphthyl-2,2’-diamine(9),aspergillusone C(10),cyclo-(DIle-L-Trp)(11),cyclopenol(12),serlyticin-A(13),communol C(14),cerebroside C(15).The antibacterial and acetylcholinesterase inhibitory activities of all the compounds were tested. The results showed that compounds 1,2,and 4 exhibited potent inhibitory activities on the tested strains, with MIC values ranging from 1 to 4 μg/m L.Compounds 3 and 8 had inhibitory effects on acetylcholinesterase, and their inhibition rates were 17.53% and 10.48%,respectively.In the third chapter, the solid fermentation product of P.crustosum JT-8 was systematically isolated and analyzed.Seventeen compounds were isolated and identified as: squalene(1),sorbicillin(2),methyl indole-3-acetate(3),indole-3-acetic acid(4),scytalone(5),6-[(2E,6E)-10,11-dihydro-11-hydroxy-farnesyl]-5,7-dihydroxy-4-methylphthalide(6),ergosterol(7),viridicatin(8),3:3’-diacetyl-7:9:7’:9’-tetrahydroxy-5:5’-dimethyldiphenylmethane(9),roquefortine C(10),penilloid A(11),trichoderone(12),3,4-dihydro-3,4,9-trihydroxy-l(2H)-naphthalenone(13),3,5-dihydrosorbicillin(14),1-O-β-D-glucopyranosyl-stigmine-4,23(27)Z-diene(15),6,8-dihydroxy-5,7-dimethylacetophenone(16),cyclo-(L-Trp-L-Phe)(17).The antibacterial and acetylcholinesterase inhibitory activities of all compounds were tested. The results showed that compounds 3,9,11,13,14,and 17 possessed inhibitory activities against the tested strains, with MIC values ranging from 1 to 512 μg/m L.Compounds 2,3,4and 6 had inhibitory effects on acetylcholinesterase, the inhibitory rates were 14.71%,27.01%,12.49% and 17.53%,respectively.
