Fluorescence probe technology is a fast detection and analysis technology.It has many advantages,such as high sensitivity,fast response,convenient operation,portable device,low cost,and no need for samples to be processed.It is widely used in many fields,such as chemistry,biomedicine,food hygiene,environmental monitoring,city safety and many other fields.Among them,fluorescence probes for trace detection of the various ions and small molecules which are closely related to the biology systems have attracted much attention.This paper is mainly to explore new fluorescent probes with good biocompatibility,high water solubility,easily modifying and non-toxic,and to lay the foundation for its application in intracellular detection and clinical diagnosis and treatment.Triphenylamine Schiff base fluorescent probe and carbon quantum dot based fluorescent probes were prepared respectively for the detection of cyanide and zinc ions and hydrogen sulfide detection in vitro,and explored the recognition mechanism by means of experiment and theory calculation,providing a scientific basis for the design of fluorescent probe for trace detection for multi ion and small molecule in vivo.1)A Schiff base probe was prepared with three anilines as fluorophore,which was used for the detection of zinc ions and cyanogen.Characterized by multi-methods as FTIR spectrum,UV-Vis spectrum,NMR,X-ray single crystal diffraction,and tested the detection performance to the two ions.The probe can respond in UV-Vis absorption and fluorescence both,and shows good selectivity.The detection limit of zinc ions is to 14 n M,and cyanide0.37μM;also we carried on the experimental study of NMR titration to verify the ion recognition mechanism.The results of intracellular test showed that the poor water solubility of the probe lead to an unobvious phenomenon on the recognition effect.2)In order to develop a better water-soluble and biocompatible fluorescent probe,a fluorescent probe for hydrogen sulfide was designed and fabricated with a yellow fluorescent carbon quantum dots which targeted to lysosomal.Studies in the previous work showed the probe having a good detection effect and biological properties.The influence of the amount of Fe3+and p H values on the fluorescence intensity of the carbon quantum dot probe 1 was systematically studied.The possible reasons were verified by experiments,which lay the foundation for subsequent application.
