Immunochromatography is a new immunoassay technology originated in the 1980 s.Later,it has been widely used in food detection,environmental protection and other fields.In recent years,the technology has been gradually applied to the medical clinical detection.It is favored by researchers for its advantages of simple operation,low cost,rapid and accurate detection.At present,nucleic acid detection and enzyme-linked immunosorbent assay are mostly used for many viruses with great harm and strong infectious ability.These two methods are complex and take a long time.It needs many equipment and instruments to complete the whole detection process,so these viruses still can not achieve the purpose of effective and rapid detection.In order to solve this kind of problem,we use the established chromatography immunoassay strip to detect the viral antigen qualitatively,semi quantitatively and quantitatively.This simple and effective method can replace the traditional detection method.The basic content of this paper can be divided into the following three parts:1.Detection of Shiga toxin type II based on colloidal goldIn this chapter,through the preparation of chromatography immunoassay strip platform and the synthesis of colloidal gold properties,we can achieve the purpose of rapid qualitative detection of Shiga toxin type II(STX2)in 10 minutes.Under the condition of only observing with eyes,according to the color change on the test(T)line,it can be judged that the detection limit is about 25 ng/ml,and the result is stable after repeated detection.The designed strip is easy to carry and has a long storage time.No need to use any instrument and equipment for testing,only need to add 50 μL standard solution,even in remote areas,it can be tested in real time.This strip provides a very simple detection method for qualitative detection of antigens in the future,and has a large-scale commercial application prospect.2.Detection of Shiga toxin type Ⅱ based on CdTe quantum dotsIn this chapter,CdTe quantum dots in water phase are replaced to detect STX2.The chromatographic immunoassay strip based on CdTe quantum dots can detect STX2 semi quantitatively.The special properties of quantum dots also make the detection limit lower than that of colloidal gold probe.Under the UV light,the detection limit can be reduced to 10ng/ml through the change of fluorescence intensity on the T-line.The establishment of this strip can realize the rapid quantitative detection of STX2 and provide a new semi quantitative method for the detection of antigens in the future.3.Detection of Zika virus based on fluorescent nano silicon spheresIn this chapter,a kind of composite material is used to form silica fluorescent nanospheres by the way that the silica spheres contain oil phase ZnCdSe / ZnS core-shell quantum dots.The reason for the quantitative analysis of Zika virus with the chromatographic immunoassay strip made of this material as a probe is that the fluorescence efficiency of the core-shell oil quantum dots is higher than that of the water quantum dots,so its application in biological detection can reduce the detection limit more.However,in order to overcome its non hydrophilic shortcomings,it is wrapped in silica,which also avoids the impact of metal ions on the environment.In the detection of Zika virus,the detection limit is as low as 100pg/ml by analyzing the captured detection images with Image J software.The enhancement of fluorescence intensity on T-line changed with the increase of antigen concentration.The best result was observed 15 minutes after the sample was dripped.The experimental results show that the strip is very specific,reproducible and stable.This proves that the immunochromatography strip platform has the reliable practical application ability.
