| Dendrocalamus latiflorus Munro is a perennial woody plant belonging to the subfamily of Gramineae Bamboo,which is widely distributed.The secondary metabolites of Dendrocalamus latiflorus Munro leaves are rich,the biological activities and pharmacological functions are diverse,and the development and application value is high.This article takes Dendrocalamus latiflorus Munro leaves in Chishui area of Guizhou Province as the research object,explores the extraction and preparation of its flavonoids and acid hydrolysis process,and extracts them The anti-oxidant activity of the product and acid hydrolysate was studied,aiming to provide a theoretical basis for the development and utilization of Chishui Dendrocalamus latiflorus Munro leaf resources.The main research contents and results are as follows:1.Study on the extraction and preparation technology of flavonoids from the leaves of Dendrocalamus latiflorus MunroOn the basis of the small test,the extraction process of the flavonoids from the leaves of Dendrocalamus latiflorus Munro was scaled up step by step.With 20-mesh sesame leaf powder as the raw material,the feed amount is enlarged from 0.1 kg to1.0 kg,and then to 20.0 kg,the extraction solvent is 90%ethanol,the extraction times are 3 times,and the material-to-liquid ratio is 1:10(Kg:L),1:7(kg:L),1:7(kg:L),the extraction time is 2 h,1.5 h,1.5 h,and the extraction temperature is 80~85℃.The results showed that the total flavonoid content of Ma Bamboo leaf extract reached over 8.28%,the yield reached over 9.68%,and the total content of orientin,isoorientin,vitexin and isovitexin reached over 0.71%;ethanol The recovery rate reaches 90%.2.Study on the Acid Hydrolysis Technology of the Extract from the Leaves of Dendrocalamuslatiflorus MunroA scale-up test of 10.0 g was carried out on the basis of the 0.1 g(feeding amount)of the acid hydrolysis test.Hydrolysis conditions are:substrate concentration1.0 mg/m L,hydrochloric acid concentration 1.2 mo L/L,reaction time 3 h,reaction temperature 95℃.The results showed that the total flavonoids content of the acid hydrolyzate of the extract of Dendrocalamus latiflorus Munro leaves reached 29.49%,and the total flavonoids content of the four carbon glycosides reached 2.25%,which were 3.51 times and 3.08 times higher than those before hydrolysis.3.Study on the ultrasound-assisted acid hydrolysis process of the extract of Dendrocalamus latiflorus Munro leavesTaking the content of four carbon glycoside flavonoids in the leaves of Dendrocalamus latiflorus Munro leaves as the detection indicators,based on the single factor test,the ultrasonic-assisted acid hydrolysis process was optimized by the response surface method.With fixed substrate concentration(1.0 mo L/L)and ultrasonic power(300 W),under the conditions of hydrochloric acid concentration of1.0 mo L/L,temperature of 65℃,and time of 147 minutes,The ultrasonic-assisted acid hydrolysis amplification test was carried out from 0.01 g to 1.0 g.The total flavonoid content of the acid hydrolyzate was 35.52%,and the total content of the four carboside flavonoids was 2.85%.4.Antioxidant activity of the extract from the leaves of Dendrocalamus latiflorus MunroThe ability to remove DPPH·,·OH and O2-·of endrocalamus latiflorus Munro leaf extract and acid hydrolysate was tested.The results showed that both the extract of endrocalamus latiflorus Munro leaf and the acid hydrolysate had certain antioxidant activity,among which:the scavenging rate of the extract on DPPH·,·OH and O2-·were 71.53%,73.01%,73.23%,respectively;the scavenging rates of acid hydrolysate to DPPH·,·OH and O2-·were 82.19%,43.68%,59.11%.In this paper,the extraction process of flavonoids in the leaves of Dendrocalamus latiflorus Munro,the acid hydrolysis process of the extract,and the ultrasonic-assisted acid hydrolysis process were systematically studied,and the antioxidant activity of the extract and acid hydrolysate was determined.The preparation of carbon glycoside flavonoids in bamboo leaves and the development of functional products provide a reference,which has important theoretical significance and application value for the comprehensive utilization of bamboo leaf resources in Chishui area. |
