The cyclic ether structure exists widely in natural products and bioactive molecules which is one of the common structural motifs in synthetic drugs.Therefore,in the development of pharmaceutical chemistry,it is of great significance to developα-functionalization of cyclic ethers to realize the synthesis of novel ethers.In this paper,the copper-catalyzed α-C-H allenylation of ethers was studied.At the same time,the recent research progress of α-functionalization of inactivated ethers and 1,4-addition of1,3-enynes were discussed.The paper consists of the following two chapters.Chapter 1:Research progress of α-functionalization of inactivated ethers and1,4-addition of 1,3-enynesThe first chapter is divided into two parts.Firstly,the research progress of α-functionalization of inactivated ethers was introduced.According to the different reaction substrates and forming bond type,the recent representative research work in this field was discussed.Secondly,the research progress of 1,4-addition of 1,3-enynes to allenes was introduced.According to the different catalysts used in the system,we discussed these reactions in detail.Chapter 2: Study on copper-catalyzed α-C-H allenylation of cyclic ethersThe α-allenylation reaction was realized through the copper-catalyzed 1,4-addition of cyclic ethers and 1,3-enynes,and the cyano group was introduced at the same time.Under mild conditions,various structures of cyclic ether derivatives,containing allene functional groups,was synthesized with moderate to good yield and high regioselectivity.The reaction showed good functional group compatibility to 1,3-enynes,but the range of substrates for ethers was limited.
