This paper presents an analytical method for simultaneous determination of three basic antihypertensive drugs using strong cation-exchange microcolumn liquid chromatography with silica particles supporting polymeric monolithic column(shortened as s-column).The s-column was obtained through first preparing a monolithic matrix in a 150μm inner diameter fused silica capillary packed with 5μm bare silica particles by polymerization using glycidyl methacrylate(GMA)as the monomer,pentaerythritol triacrylate(PETA)as the crosslinking agent,a mixed solution of methanol and cyclohexanol as the porogen,and2,2-azobisisobutyronitrile(AIBN)as the initiator,and then the monolithic matrix was bonded the group of-SO3H by the reaction of the epoxy active group in GMA and sodium sulfite solution.A micro-column liquid chromatographic analytical method was established for simultaneous determination three basic antihypertensive drugs in human plasma and urine using UV as detector.The three antihypertensive drugs are atenolol,hydrochlorothiazide and reserpine.Firstly,the experimental conditions,including mobile phase p H,the phosphate and the organic additive concentrations in the mobile phase,were selected for the chromatographic separation.Then some key values for evaluation of analytical method were measured.The detection limits are lower than 0.30μg/m L,the intra-day and inter-day precisions are no more than 5.0%,and the recoveries are higher than 90.0%for the three drugs in plasma and urine;and the linear ranges are 2.0-16.0μg/m L,1.0-16.0μg/m L and0.5-16.0μg/m L for HCT,ATN and RSP in plasma,respectively,and 2.0-24.0μg/m L,2.0-24.0μg/m L and 1.0-24.0μg/m L for HCT,ATN and RSP in urine,respectively.
