Objective To prepare liposomes of egg yolk antibodies against Porphyromonas gingivalis(anti-Pg-IgY liposome),and to study the antibacterial effect of these anti-Pg-IgY liposomes on Porphyromonas gingivalis,a major periodontal pathogen,as well as to observe the effect of these liposomes on experimental periodontitis in rats.Design : This study aimed to develop safe and effective biological preparations of yolk antibodies specific to the main pathogenic bacteria of periodontitis using yolk antibody technology and nanotechnology.Anti-Pg-IgY liposomes were prepared by film ultrasonic dispersion method.The appearance of anti-Pg-IgY liposomes was observed by transmission electron microscope.Zeta potential and particle size of anti-Pg-IgY liposomes were measured by dynamic light scattering nanoparticle size analyzer.The minimal inhibitory concentration(MIC)of anti-Pg-IgY liposomes against Porphyromonas gingivalis was studied by microdilution method.Physical,morphological and ultrastructural analyses were performed to evaluate the drug entrapment efficiency,drug release curve and antibacterial activity in vitro.In addition,30 rats were divided into three groups: anti-Pg-IgY liposome group,minocycline group,anti-Pg-IgY solution group,PBS group,control group and blank group.The weight,probing depth(BOP),gingival bleeding index(SBI),tooth mobility(mob)and other periodontal clinical indicators were recorded during the modeling and treatment.After 8weeks of treatment,the rats were killed,and their mandibles were removed for micro CT analysis and he histological staining.The data of each group were analyzed by paired t test and SPSS 22.0.Results :The average particle size of the liposomes was 175.43.±3.25.nm(n = 4),while the polydispersity index was 0.263(n = 4),and the zeta potential was-37.82±0.20.mV(n = 4).The encapsulation efficiency was 42.71%(n = 4),and the cumulative release rate was 83.4%.The minimum inhibitory concentration of anti-Pg-IgY liposome to P.gingivalis was 4 mg / ml.Periodontal parameters,including MOB,PD,and SBI,were significantly improved in both the anti-Pg-IgY liposomes and minocycline hydrochloride groups at the end of treatment(P < 0.05).There was no significant difference in PBS buffer group before and after treatment(P > 0.05).Histological observation showed that anti-Pg-IgY liposomes could control local inflammation and repair tissue.Conclusions The results confirmed that the anti-Pg-IgY liposomes exhibited excellent antibacterial activity and successfully promoted periodontal tissue repair,making them a promising therapeutic agent for the treatment of periodontitis.
