| Since the 1960 s,a large number of organic ligands have been found to have positive anti-cancer therapeutic effects,and a large number of metal complexes have been synthesized and their anti-tumor activities have been studied.Transition metals Co,Cu,Fe,Zn,Mn,Ni,Cr,etc.are trace elements necessary for the human body,which are necessary to maintain the body’s metabolism.When selecting metals for the synthesis of complexes,transition metals have become the preferred element due to their particularity to the human body.The complexes exhibit different biological activities due to their different coordination modes,so the choice of ligand has a huge impact on the final anti-cancer activity of the complex.2-(1,2,4triazolyl)-isonicotinic acid has abundant coordination sites and good coordination ability,and the azole group has the characteristics of good biological activity,so it is chosen as the ligand.In this paper,three complexes of Co(II),Cu(II)and Zn(II)were synthesized by hydrothermal method,and their structures were characterized.The specific research contents are as follows:1.Three kinds of complexes(1-3)are synthesized by hydrothermal method,and the data is collected by X-ray single crystal diffractometer for structural analysis,and the complex structure and substance purity are verified by infrared spectroscopy and X-ray diffractometer.2.Fluorescence spectroscopy is used to study the binding mode of complexes 1-3 and CT-DNA.The Kb values of the three complexes and CT-DNA are the results of computer testing and calculation through ultraviolet experiments.Two spectra The experimental results show that the complexes 1-3 all interact with DNA in an insertion manner.The ability of the complex to cut p BR322 plasmid DNA was tested by gel electrophoresis.3.Use flow cytometry to detect the apoptosis ratio of complexes 1-3 to He La cells,and compare the strength of the complexes to induce apoptosis in He La cells.With the aid of a fluorescent inverted microscope,the deformation state of the cells was observed by Annexin-PI double staining method to compare the apoptosis effect of complexes 1-3 on He La cells.The above results indicate that the copper complex has the strongest induction of apoptosis on HeLa. |
