In the physiological environment of the organism,cysteine(Cys),homocysteine,(Hcy),glutathione(GSH),sulfur dioxide(SO2),N-acetylcysteine(NAC)and other active sulfur substances have played a very important and crucial role.In addition to SO2 and H2S,many bio thiols such as Cys,Hcy,GSH and NAC have similar chemical constitution and characters,but their concentration and biological functions are quite different in biological cells or blood serum.In addition,various biological thiols are interrelated.Sulfur dioxide(SO2)is an important metabolite of GSH and Cys,which is usually related to symptoms of nervous system diseases,cardiovascular diseases and lung cancer.Therefore,as a simple and reliable detection tool,the fluorescent probe with advantageous function can monitor the production,change,flow,concentration change and metabolic process of these biological thiols in actual time,which is very meaningful for the study of certain physiological activities and related of diseases.The primary coverage of this paper are as below.1.A fluorescent probe X for monitoring SO2 was designed and synthesized.Based on the nucleophilic addition reaction between SO2 andα,β-unsaturated carbon-carbon double bonds,probe X can quickly and sensitively detect SO2 derivatives,emit 488 nm green light at an excitation wavelength of 410 nm,and is successfully used for fluorescence imaging analysis of SO2 in living cells.2.A dual-signal fluorescent probe M with dual reaction sites was designed and synthesized to distinguish Cys,Hcy/GSH/NAC at the same time.It can distinguish Cys(λexem=360/450 nm),Hcy/GSH/NAC(λexem=460/536 nm)at the same time,which is successfully used for distinguishing exogenous and endogenous Cys,Hcy/GSH/NAC and fluorescence imaging analysis at the same time in cells.3.A multi-signal fluorescent probe H was designed and synthesized to simultaneously discover endogenous and exogenous Cys,SO2,GSH and Hcy in cells and zebrafish.Probe H uses coumarin as the fluorophore and contains 4 active reaction sites with different spatial distances,which achieves high sensitivity and selectivity while distinguishing Cys(λexem=367/464 nm),SO2exem=425/520 nm),GSH(λexem=450/536 nm)and Hcy(λexem=490/564 nm).Probe H successfully used to simultaneously visualize endogenous and exogenous Cys,SO2,GSH and Hcy and fluorescence imaging analysis in cells and zebrafish.
