With the development of the aging of the population,the health problems caused by aging will undoubtedly bring great burdens to society.Free-radical theory is an important theory about aging in recent years.The free-radical theory believes that the core factor of aging is oxidative damage,which is caused by reactive oxygen species(ROS).The high level of ROS in cells can lead to damage such as DNA damage,unfolded protein responses,and cell signaling disturbances.The long-term accumulation of oxidative damage eventually leads to cell senescence,apoptosis,etc.At the macroscopic level,oxidative damage also leads to body aging.The Chinese traditional herb longan arillus is one of the herbs of C hinese medicinal and food homologous list,which is dried from the fruit longan.The medicine book “Shennong Materia Medica” has a record of use: long-term use can enhance the soul,make people smart,strong,long-lived,and theosophical.This description meets the standards of modern medicine against aging.Therefore,this C hinese medic ine was selected as the research object in this study.Caenorhabditis elegans is a classic model organism in the studies of lifespan.C.elegans has homologous genes of many human diseases.Besides,C.elegans has lots of advantages,such as a short lifespan,small size and easy observation.Therefore,based on the separation and extraction of longan meat in different ways,this paper explored the effects of longan arillus components extracted by different ways on the antioxidant damage ability in C.elegans.In this study,ultraviolet radiation was used as a stress source,and C.elegans was used as a model organism.Basing on the Free-radical theory of aging,the ability of traditional C hinese herb longan arillus on resist oxidative damage was explored.After ensuring that it would not cause caloric restriction to C.elegans,anti-stress and life tests were carried out on different dosing times and different drug extraction methods.As a result,both water extract and alcohol extract from ongan arillus could slightly prolong their life span and significantly prolong their lifespan after stress.However,ongan arillus had preventive effect as feeding ongan arillus after stress had negative effect on C.elegans.After that,the anti-stress effect of longan arillus was determined by the ROS content in C.elegan,which was determined by the upregulation of the translocation of forkhead protein DAF-16.Then,the expression of some downstream genes of DAF-16,such as was detected.It was found that the expressions of hsp16.1,hsp16.2,hsp-12.6 and sod-3 genes were significantly up-regulated.Then,the water extract of ongan arillus was extracted by ethanol,ethyl acetate,petroleum ether and n-butanol.The effect of ethanol phase and ethyl acetate phase could almost reproduced the effect of water extractbe,while the effect of petroleum ether phase was poor.Polyphenols were the main components in ethanol phase and ethyl acetate phase,and phospholipids were the main components in petroleum ether phase.Therefore,the effective components of anti-stress in ongan arillus aqueous were mainly polyphenols.Finally,the content of flavonoids with strong antioxidant capacity and polyphenols was further determined,and the dose effect relationship was also determined.In summary,this paper confirmed that the water extract and alcohol extract of ongan arillus had a good effect on prolonging the life of nematodes and resisting UV radiation stress.The mechanism of delaying aging of water extract from ongan arilluswere further explored: by promoting the nuclear translocation of DAF-16 in insulin-like growth factor signaling pathway and regulating the expression of downstream target genes.As a result,the content of reactive oxygen species in nematodes was reduced.Finally,the polyphenols in longan arillus were identified as the main active ingredient.Therefore,the study can provide some new ideas for the research and development of drugs against oxidative stress,such as the method of activating DAF-16 mediated longevity mechanism,and the mechanism of drug sensitivity difference of C.elegans in different life stages.In addition,this paper hopes to promote the development of longan and deep-processing industry of ongan aril us.
