This thesis included following four parts:(1)Chirality and chiral compounds were introduced.The different enantioselective resolution methods such as membrane separation and high performance liquid chromatography etc.were mentioned.The characteristic of phenylglycine was stated.(2)The vancomycin composite membrane of 144 cm~2was prepared to separate phenylglycine.The effects of feeding concentration,temperature,time and p H were optimized.The orthogonal experiment of L9(3~4)was designed.The maximum enantiomeric excess value(e.e.)could reach 92.0%,which the successive dialysis was carried out to obtain 95.0%e.e.(3)The preparation of 144cm~2cyclodextrin composite membrane for chiral separation of phenylglycine was studied.When the feeding concentration was 0.1mg/m L,the temperature was 30℃,the dialysis time was 1d,and the p H was 7.0,over 84.5%enantiomeric excess value could reach.During secondary dialysis 90.0%e.e.dialysis result could be shown.(4)We also crossly linked the sodium alginate membrane with glutaraldehyde to extract the phenylglycine.The orthogonal experiment of L9(3~5)was designed.The maximum enantiomeric excess value could reach 61.3%under the condition of0.04 mg/m L phenylglycine,p H 7.0 and extracting 20 min at 25℃.During the second extraction,the extraction was able to give 77.0%e.e.The above experiments all show that the three different composite membranes have good recognition ability and separation effect when exploring the separation of D,L-phenylglycine,and the vancomycin composite membrane has the best separation effect;Significance and development value of chiral drug separation. |
