·ObjectiveIn the reassarch,we adopt the matebolomic technology to study the characteristic metabolites and their differences、variation tendency in each hypeiuricemia model rat groups.·MethodsIn the experimental animal center of Zhejiang Chinese Medicine Univ ersity,we give the rats uricase inhibitor and high purine diet to establish the hypeiuricemia rat model.We use biochemistry detection technology to verify the hypeiuricemia rat model have been established successfully,then we take the blood of all rats to get the serum by the highspeed separatio n.First,using the automatic biochemical analyzer to test the biopsy,thenus ing the metabonomics technology to detect the small molecule metabolites in serum.Next,we use metabolites fingerprint to obtain subsection integr-a 1.After the normalization processing of integral data,through Principal Com ponent Analysis and Partial Least Squares analysis to get scoring diagram and the load diagram.According to the two diagrams,We find out the diff-erences of characteristics metabolites to identify the biomarkers and its or igin and metabolic pathways and distinguish metabolic profile and find po-tential biomarkers and metabolic pathways.ResultsThrough the results of Principal Component Analysis model,we are in formed of that the blank group and model group can be completely separ-ated and the model+allopurinol group and model group can be observed obvious clustering and approximately separate by Key metabolic product,as well as there is trend of transition to a model group.From the PLS-D Achart and the load diagram analysis,metabolites of blank group,model g-roup and the model+allopurino group do exist differences.Comparing with the blank group,the plasma of model group contains lower proline、proline、tryptophan、alloisoleucine、phenylalanine(P<0.01)statistically significant;In the blood of model group,the content of Acetyl Co-A、dehydrocholester-ol、cysteine is lower than normal blood(P<0.01)statistically significant.Compared with the blank group,the contents of cool base glycine,cardioli pin,glycerol phospholipid,scabb-ard glycolipids,phospholipids,inositol phosp holipids and ceramide are higher in the blood of the model group and m odel+allopurino group.The result have statistical meaning(P<0.01).On th ebasis of these results,both model group and model+allopurinol grou-p ra tsallhave lipid metabolism,glucose metabolism,protein metabolism abnorm alities or disorder.These changes may be a bridge role in the process of evolution of metabolic syndrome.These differences among blank group,mod el group and blank+allopurinol group are about lipid,amino acid,lactic acid and other substances,this is also consistent with the concept of the m-etab olic syndrome.And the material is the inevitable result of the body carboh-ydrates,carbohydrate,lipid,protein metabolic disorder.ConclusionHigh uric acid level in model rats body has affected the metabolism and metabolic outcomes of lipid and protein,at the same time also led to the body energy metabolism and immune system disorders.And between di-fferent experimental groups,different degrees of change are happened in different metabolism or metabolic networks.
