| Aging is an inescapable and irreversible destiny of mammals.The theories of aging occurrence are confirmable in aging and aging-related diseases.More and more theoretical and experimental foundations support that cellular senescence will induce functional degeneration of tissues and organs.Nowadays,the role of nutrients in anti-aging has drawn increasing attention with the advances in our living standards.Here we focused on the effects of 20 amino acids and common sugar on cellular senescence,and then tried to study the underlying mechanisms of Lactose and Arginine.Lactose was a common nutrient in daily life,originally found in milk and milk products.In present study,we revealed Lactose could induce the expression of SA-β-gal and p16ink4a,both were the characteristics of cellular senescence,as well as increase intracellular oxidative stress by regulating Nrf2 pathway to accelerate senescence.In addition,Lactose treatment could shorten the lifespan of C.elegans,which was rescued by antioxidant NAC.L-Arginine was a non-essential amino acid to human.The main source of plasma Arginine was dietary intake besides auto synthesis.In our study,the effects of Arginine on MRC-5 cell lines cultured under high glucose were examined.It was found that Arginine could increase eNOS activity effectively via activating PI3K/Akt/eNOS pathway,which ultimately resulted in the increase of NO production and resistance to cellular senescence induced by high glucose.In summary,we set up the aging models in vitro and investigated the effects of common nutrients on cellular senescence and their mechanisms,which provided the experimental foundation for our future anti-aging study. |
