Objective:Investigate the expressions of hypoxia-induced factor-1α(HIF-1α)and heat shock protein(HSP70)in hepatic ischemia reperfusion injury(HIRI)in mice,and to explore the mechanism of hif-1α expression and HSP70 in the pathophysiological process of HIRI.Methods:60 SD rats were randomly divided into sham group(S group),ischemia-reperfusion group with the 3 hour reperfusion(I/R 3h group)and ischemia-reperfusion group with the 6 hour reperfusion(I/R 6h group),liver ischemia-reperfusion model was established by Pringle maneuver.Immunohistochemistry(IHC)method was used to detect the expression of HIF – 1α and HSP70 in S group,I/R 3h group and I/R 6h group.Histopathological observation were carried out under the optical microscope and semi-quantitative analysis was chosen to assess the results.Results: 1.HIF-1α expressed in hepatocyte nucleus and cytoplasm in S group,I/R 3h group and I/R 6h group,mainly in the nucleus.The positive expression rate of HIF-1α in 3 groups was statistically significant(χ2 =19.564,P<0.05).The positive expression rates of I/R 3h group and I/R 6h group were higher than those of S group(P<0.05).The positive expression rate of hepatocytes in I/R 6h group was higher than that in I/R 3h group(P<0.05).2.HSP70 expressed in hepatocyte nucleus and cytoplasm of S group,I/R 3h group and I/R 6h group,mainly in the nucleus.The positive expression rate of HSP70 in 3 groups was statistically significant(χ2 =13.923,P<0.05).The positive expression rates of I/R 3h group and I/R 6h group were higher than those of S group(P>0.05).There was no statistical significance in the positive expression rate between the I/R 3h group and the I/R 6h group(P>0.05),and the positive expression intensity of the I/R 6h group was higher than that of the I/R 3h group(P<0.05).3.In the liver tissues of the I/R 3h group and the I/R 6h group,the expression of HIF-1α was positively correlated with the expression of HSP70,and the correlation coefficients were 0.460(P<0.05)and 0.593(P<0.05),respectively.Conclusion: 1.The expression of HIF-1α in hepatic ischemia-reperfusion injury was enhanced,and the expression was also enhanced with the aggravation of the injury degree.2.The expression of HSP70 in hepatic ischemia-reperfusion injury was enhanced,and the expression was also enhanced with the aggravation of the injury degree.3.There was a positive correlation between HIF-1α and HSP70 expression in liver tissue after hepatic ischemia-reperfusion injury,suggesting that HIF-1α and HSP70 were involved in the pathophysiological process of hepatic ischemia-reperfusion injury.
