Objective:To explore the application of Optical Coherence Tomography OCT in children with Neurofibromatosis type1 NF1 combined with Congenital pseudarthrosis of the tibia CPT.Label Free Quantitation was used to analyze the differential expression of proteins in the tibia of children with NF1 combined with CPT and non-NF1 combined with CPT induced labor to explore the possible pathogenesis of NF1 combined with CPT.MethodsTen children with NF1 and CPT who were treated in our hospital were selected as the experimental group.OCT scan and routine pathological staining were performed after tibial tissue specimens were obtained,and the results of the two were compared.The tibias of 5 non-NF1 non-CPT induced fetuses were selected for the above examination as the control group,and the results of OCT scan and routine pathological staining were compared in each group.Randomly selected 3 cases of NF1 combined with CPT diseased tibia and 3cases of non-NF1 combined with CPT induced labor tibia for comparative analysis,using non-labeled quantitative Tandem Mass Spectrometry Technique(Liquid Chromatography Tandem Mass Spectrometry Technique,LC-MS/ MS)technology compares and analyzes the two sets of samples to obtain 1508 Significantly Differentially Expressed Proteins(SDEPs),including 516 up-regulated proteins and992 down-regulated proteins.The Max Quant database and Uniport database are used for analysis and bioinformatics analysis.ResultsOCT scan showed scattered blood vessels,fats and nerves in the specimen.OCT scans of the control group showed dense shadows.The experimental group saw disordered and highly refractive osteoclasts,scattered blood vessels,vacuoles of fat and nerves..SDEPs are mainly involved in the complement and coagulation cascade,protein post-translational modification,the regulation of bone healing,and the regulation of the PI3K-AKt signaling pathway.These pathways may play an important role in the pathogenesis of NF1 combined with CPT.ConclusionOCT directly scans freshly isolated diseased tissue,and then presents the diseased structure inside the tissue,which has a high degree of overlap with the imaging examination under the same stage of the optical microscope;OCT supplements the histopathological diagnosis of NF1 combined with CPT,and is also a further gene Keep samples for academic inspection.There are differential expression of proteins in the tibia of children with NF1 and CPT and the tibia of non-NF1 with CPT.The metabolic pathways and pathways involved in the regulation of these differentially expressed proteins may play an important role in the pathology of neurofibroma and the pathology of tibial pseudoarthritis.
